Purpose:Through the traditional Chinese medicine,"Yi Tang Kang," study theeffect of urinary protein, angiotensin Ⅱ, endothelin-1expression and thechange of the structure of renal tissue in rats with diabetic nephropathy,to be proved that traditional Chinese medicine,"Yi Tang Kang" for diabeticnephropathy rat kidney has a protective effect, can delay the occurrence ofkidney damage.Material and method:The first60SPF SD adaptive feeding rats a week.Randomlyselected10rats as the blank group,the remaining50rats were fed with highfat diet and intraperitoneal injection of streptozotocin (STZ) method formodeling.The successful model of the50rats were randomly divided into modelgroup,prevention group,Yi Tang Kang high dose group,Yi Tang Kang low dosegroup,Benazepril group.Prevention group in diabetic models after given medicineyitangkang standard dose administered.The remaining four groups after modelingin accordance with their respective processing factors for stomach,Model groupwere given distilled water. In the administration of sixth weekend metaboliccage of rats in each group were collected24hours urine.Finally, remove thekidney. Determination of each24hour urinary protein quantitative by coomassieblue staining。Determination of Ang Ⅱ in renal tissue by enzyme linkedimmunosorbent assay(ELISA),tection of renal tissues of rats in each group theexpression of ET-1mRNAby reverse transcription PCRof ET-1mRNA, Detection ofendothelin ET-1protein expression by Western blot and Observing pathologicalchanges.Results:1.The Blood glucose value of model group was significantly higher than blankcontrol group (P<0.01);Compared with the model group, Chinese medicineprevention group, low-dose group, high dose group, western medicine group, Blood glucose value were significantly reduced (P<0.01),no significant difference oftraditional Chinese medicine prevention group, high dose group and Westernmedicine group (Table1).Explanation: Yi Tang Kang can significantly reduce thecontent of Blood glucose value. Benazepril had no hypoglycemic effect. The24hour urine protein of model group was significantly higher than blank controlgroup (P<0.01),Compared with the model group, Chinese medicine prevention group,low-dose group, high dose group, western medicine group,24hour urine proteinwere significantly reduced (P<0.01), difference between traditional Chinesemedicine low-dose group and Western medicine group.(Table1). Explanation: YiTang Kang can significantly reduce the content of24hour urine protein.2.The AngII of model group was significantly higher than blank control group(P<0.01);Compared with the model group, Chinese medicine prevention group,low-dose group, high dose group, western medicine group, AngII weresignificantly reduced (P<0.01),no significant difference of traditional Chinesemedicine prevention group, high dose group and Western medicine group (Table2).Explanation: Diabetic nephropathy state, local kidney RAS isactivated,abnormal increase of AngII content in kidney.Yi Tang Kang cansignificantly reduce the content of AngII in kidney tissue.3. The ET-1protein expression of model group was significantly higher than blankcontrol group (P<0.01);Compared with the model group, Chinese medicineprevention group, low-dose group, high dose group, western medicine group, ET-1protein expression were significantly reduced (P<0.01), no significantdifference of traditional Chinese medicine prevention group, high dose groupand Western medicine group (Table3). Yi Tang Kang can significantly reduce thecontent of ET-1protein expression in kidney tissue.4.The ET-1mRNA of model group was significantly higher than blank control group(P<0.01),Compared with the model group, Chinese medicine prevention group,low-dose group, high dose group, western medicine group, ET-1mRNA weresignificantly reduced (P<0.01),no significant difference of traditional Chinesemedicine prevention group, high dose group and Western medicine group (Table 4).Explanation: Diabetic nephropathy state, local kidney ET isactivated,abnormal increase of ET-1mRNA content in kidney.Yi Tang Kang cansignificantly reduce the content of ET-1mRNA in kidney tissue.Conclusion:1.Diabetic nephropathy state, local kidney RAS and ET is activated. Both togetherlead to increased renal damage in diabetic nephropathy.2.Yi Tang Kang can be effectively reduced renal tissue angiotensin Ⅱ andendothelin expression. Has a role in protecting the kidneys.3. Early use of Yi Tang Kang intervention on diabetes, which can delay theprogress of renal damage.
