Antibiotics for various types of bacteria, fungi, viruses and mycoplasma infections with different degrees of preventive and therapeutic effects, has a wide range of clinical applications. Human serum albumin which is capable of binding a great variety of metabolites and drugs is the most abundant carrier protein in blood plasma. Antibiotics combine with HSA in varying degrees into the blood and the binding will significantly influence the absorption, distribution, metabolism excretion, stability and toxicity of the drugs. Therefore, investigating the interaction of antibiotics with HSA are very important for the study of pharmacokinetics and toxicology, and even influence the research and development of new drugs.In this paper, the interactions between antibiotics (include levofloxacin, miconazole nitrate, minocycline, sulfadiazine, cefazolin and ceftriaxone) and HSA in physiological solution were studied by multi-spectroscopic methods and molecular docking methods. The inner filter effect was corrected in order to obtain accurate data. The binding parameters such as binding constant, the number of binding sites, thermodynamic parameters and the binding distance between Trp214 of HSA and the antibiotics were calculated by used of the Stern-Volmer equation, the Vant’s Hoff equation and the Forster theory on resonance energy transfer. The three dimensional fluorescence and synchronous fluorescence spectra indicated the conformation of HSA changed after adding the antibiotics, and the alternation of protein secondary structure were quantitatively calculated from FT-IR and CD spectra. Furthermore, the binding details between the antibiotics and HSA were further confirmed though molecular docking methods. Moreover, the interactions between LVFX, MNC with HSA were studied by’H NMR.
