Objective:Explore the relationship among the tonify deficiency and remove blood stasis prescription chanshu granule, estrogen and ERK1/2 signaling pathway, detect the level of serum estrogen, Phosphory Cyclic-AMP response element binding protein(p-CREB), Phosphory extracellular signal-regulated kinase (p-ERK1/2),Methods: 1)The Sprague Dawley rats were ovariectomized bilaterally and hormone(estrogen,progestin)injected subcutaneously for the model of postpartum depression. 2)Intervented the rats with different concentration(blank,control, shenguiren high dose, low dose, and sertraline hdrochloride) 3)Analysed the level of serum estrogen, phosphory CREB,ERK1/2 in frotal lobe of brine by western blot,Results: 1) Forced swim test showed that there is a statistically difference between blank and control(P<0.05). According to the result of estrogen,the level of control is lower than the blank,shenguiren high dose, low dose and sertraline hydrochloride. About the result of p-CREB in frotal lobe of brine, compared with the control shenguiren hige dose, low dose and sertraline hydrochloride is significantly lower than it(P<0.01). The expression of p-ERK1/2, control group is lower than the blank shenguiren low dose, and sertraline hydrochloride(P<0.01).Conclusion: 1) The model of postpartum depression can be steadily founded by ovariectomized bilaterally and hormone (estrogen,progestin) injected subcutaneously.2) Estrogen may mediate the treatment of postpartum depression by tonify deficiency and remove blood stasis.3) The antidepressant effect of shenguirenheji probably by increasing the levels of phosphory CREB, phosphory ERK1/2 in frotal lobe of brine.
