PURPOSE:Head looking for the composition of Mu an eye ointment prescription best honey concentration, the development of rational dosage regimen for the clinical treatment of herpes simplex keratitis (HSK) can provide an anti-virus, but also increase the corneal repair, healing and regeneration ability of side effects and provide the basis for a new type of eye ointment.METHODS:Select 28 healthy New Zealand white rabbits, using corneal scratches law model, eyes inoculated herpes simplex virus (HSV-1), were randomly divided into 7 groups, respectively, for the honey to give a concentration of 0%,3%,5%,7% of the An eye ointment head group, positive eye ointment (ACV eye ointment sold) group, the gel matrix group (blank control group) and model group (negative control group),3 times every day, a total of 15 days. Slit lamp to observe and score the record straight. After administration, the first 9 days, from 7 groups taken separately a rabbit, check cornea sections were observed using transmission electron microscopy the ultrastructure of corneal epithelium. The first 10 days after administration tear check for virus isolation to determine the titer, examining the head of different concentrations of honey-an eye on virus inhibition; through the single dose, containing 5% honey and honey containing 0% of the heads of an eye rabbit eye to transfer dynamics experiment:at different times from rabbit aqueous humor, aqueous HPLC method with acyclovir content, set up the mathematical model, through the Mathematics and Statistics extraction handle pharmacokinetic parameters were compared parameter differences; through the cornea in vitro release experiment to study honey containing 0%,3%,5%,7% of the heads of an eye through the cornea in vitro the speed and quantity:at different times from the cornea in vitro release medium, high efficiency Measuring the release of liquid medium acyclovir content, search acyclovir in vitro release kinetics of a function, calculating the speed of release parameters and the cumulative release percentage and error range.RESULTS:Slit-lamp observation showed score:9 day after administration of honey content is 5%,7% of the heads of an eye ointment group and other groups a significant difference in efficacy (score significantly different, P<0.05), between the two groups no statistically significant difference (P> 0.05), one of 5% honey group effect than 7% honey group more quickly. The results showed that virus titer,5% honey group the strongest inhibitory effect on the virus (the largest inhibition index), transmission electron microscopy observation also shows that films of 5% honey group keratopathy minimum degree of damage, repair of corneal strongest. Release in vitro corneal penetration results showed:0% honey content of the head of the release curve of an eye in line with the Peal-reed model, t50% to 1.896 hours,5 hours cumulative release rate of 0.2302%; 3% honey content in head-an eye Cream of the release curve in line with the Weibull model, t50% to 3.006 hours,5 hours cumulative release rate of 0.1562%; 5% honey content head of the release curve of an eye in line with the Peal-reed model, t50% to 1.320 hours,5 hours cumulative release rate of 0.3261%,7% honey content head of the release curve of an eye in line with the Weibull model, t50% to 2.686 hours,5 hours cumulative release rate of 0.1749 percent. 5% honey content and 0% honey content in an eye ointment at the head of transport in rabbit eyes are two-compartment model,5% honey content eye pharmacokinetic parameters:absorption half-life of 167.7min, distribution half-life of 114.7min, the elimination of half-life of 72.88min, peak time for 103.04min, peak concentration of 0.2386μg/mL, AUC for 42.78μg/ml·min.0% honey content eye pharmacokinetic parameters: absorption half-life of 72.88min, distribution half-life of 54.10min, the elimination half-life of 98.58min, time to peak for the 75.51min, peak concentration of 0.2041μg/mL(?)AUC for 30.40μg/ml·min.CONCLUSIONS:Containing 5% honey Mu an eye ointment group more effective than other groups, and its inhibitory effect on the virus is also the strongest. Containing 5% honey Mu an eye through the cornea in vitro release the fastest, honey eye ointment 3% are 2.28 times 7% honey eye are 2.03 times, with 0% honey eye the difference was not statistically significant. Containing 5% honey Mu an eye through the cornea in vitro cumulative release of the largest honey are 0% eye ointment 1.42-fold, 3% honey eye are 2.09-fold,7% are 1.86 times eye honey.5% honey group pharmacokinetic parameters showed that water in the room the longest half-life, peak concentration of the highest, the largest biology degrees.
