Viral diseases are the major factors for inducing the reproductive failure disease of pigs, in which porcine pseudorabies, parvovirus infection and Japanese encephalitis can cause abortion, stillborth or mummified body of pregnant sow. This study was designed to proceed from the actual production, we prepared a multivalent inactivated vaccine for the three reproductive failure disease, intend to simplify the immunization procedure,reduce the cost of immunization,reduce immune stimulation. The preparation of vaccine and the test results are as follows:1.The preparation of trivalent inactivated vaccine8000mL PRV with TCID50 of 10-65/0.1mL and 8000mL PPV with hemagglutinating antibody(HA) of 1:512 were harvested by infecting BHK-21 cells and IBRS-2 cells respectively. Mix the inactivated virus solution which had been enriched by dialysis and ultrafiltration with equal volumes of Japanese encephalitis virus and a certain proportion of adjuvant and emulsionize them by organization stamp mill and colloid mill. Two batches of trivalent inactivated vaccine against porcine pseudorabies, parvovirus infection and Japanese encephalitis were preparated, one batch was 1000mL and the other was 3000mL2.The test of trivalent inactivated vaccinePhysical properties and bacterial test of the trial vaccine were in line with the vaccine test. The test animals(pregnant sows, gilts and BALB/c mice) showed no abnormal reaction after inoculated with the vaccine and pregnant sows showed no abortion, stillbirth, or records of mummified. The protective immunity test in mice showed the titer of PRV neutralizing antibody reached an average of 1:28.5 and the protection rate resched 90% in mice when been attacked with 105.0TCID50 pig pseudorabies virulent strain in 21 days after the second immunization. The protective immunity test in the Kunming mice showed 80% rate of protection when been attacked with 105.84PFU Japanese encephalitis virus of P3 strain in 21 days after the second immunization. The Immunogenicity test in guinea pigs showed that the hemagglutination inhibition antibody titers against PPV can keep in 1:1024 for 10 weeks after the second vaccination.3.The Immunogenicity test of trivalent inactivated vaccine in pigs6 months gilts were vaccinated twice by intramuscular injection (with 3 weeks interval) with trivalent inactivated vaccines. The ELISA antibody titers against PRV produced by gilts afer vaccinating corresponded to the porcine Pseudorabies inactivation vaccine, the hemagglutination inhibition antibody titers against porcine Parvovirus outstripped porcine parvovirus inactivation vaccine, but the ELISA antibody against Japanese Encephalitis was lower than the Japanese Encephalitis live vaccine.
