A high-yielding strain G9-21 was obtained from G9 strain of spinosyn producer Saccharopolyspora spinosa by stable experimentation. G9-21 strain was mutated by four methods UV, Streptomycin, genome shuffling and bacitracin. The results showed that four mutation methods could enhance the fermentation ability of strains. The chemical titer of the four high-yielding mutant strains UV67, S26, R61, B13 were increased by 15.7%,21.9%,94.7%,7.3% respectively compared with their parent strains.Taking strains G9-21(Spc+Str-) and S26(Spc-Str+) as two parents, carriy on the breeding method genome shuffling. After eight rounds fuses continuously, compute the fusion rate. The result shows that the fusion rate was no longer changed after the 6th round fusion, namely for Saccharopolyspora spinosa effective fusion round is six rounds.Taking Saccharopolyspora spinosa G9-21(Spc+Str-) and Streptomyces avermitllis Z1-106(Spc=Str+) as the two parents, carry on the fusion between two different genus, for the question that it's sensitive to the shearing force of Saccharopolyspora spinosa.This experiment is to simulate the shearing force of industrial fermentation using beadings of the diameter 3mm, even size, and the 267g weight of a thousand. With the study in three aspects, which is the different quantity the headings joined, the different time the beading joined and the different time the beading taked out, the relationship between the fermentation ability of Saccharopolyspora spinosa and the shearing force is drawed a preliminary conclusion. If the shearing force is joined at the logarithmic phag of the mycelium, it'll cause the worst damage and reduce the abilily of fermentation.
