| A stable and high-yielding strain Co329-21 was obtained from Co329 strain of spinosyn producer Saccharopolyspora spinosa by stable experimentation. Co329-21 strain was mutated by five methods UV, NTG, 60Co-γirradiation, Streptomycin Sulfate and protoplast-UV. The results showed that five mutation methods could enhance the fermentation ability of trains. The chemical titer of the five high-yielding mutant strains UV125,N263,Co305,PU446,S119 were increased by 9.7%, 8.6%, 21.5%, 18.7%, 11.4% respectively compared with their parent strains.Based on S119 and PU131, the cultivation time, glycine concentration, bacteriolysin concentration, time, temperature to protoplasm release was examined. The optimal condition of protoplast release was as follows: glycine concentration 0.4mg/mL in seed fluid, cultivate 48 hours; 4mg/mL bacteriolysin in 32℃, 90 minutes. We also screened the Gao's No. 1 culture medium as the best regeneration culture medium. Finally, we obtained a strain R287 by fusing un-activated protoplast of strains S119 and PU131. Subculture test indicated that UV125, N263, Co305, PU446, S119 had relatively stable hereditary characteristics.Lastly, we checked the concentration of glucose, NH4+, soluble phosphate in fermentation fluid. They could provide the reference for large-scale fermentation of spinosads. |
