Study On Hybrid Progenies Crossed By Agropyron Dlaucum And Octoploid Trititrigia Thtough Mollecular Cytogenetic Analysis

Elytrigia (Elytrigia Desv) as a wild relative of wheat is an enormous and potential gene pool for wheat breeding. We introduced the genome of Thinopyrum into common wheat, and formed the new intermediate type of octoploid Trititrigia.Octoploid Trititrigia had ABD genome of common wheat and one genome ofThinopyrum, it was contained the genome of Thinopyrum intermedium, butlacking perennial genes from Thinopyrum chromosomes. This paper first usedMaicao 8, Maicao 9,and Yuanzhong 2 of Octoploid Trititrigia to hybrid withThinopyrum intermedium and obtained intergeneric hybrids, which the genes ofThinopyrum contained perennial character and strong winterness were introducedinto octoploid Trititrigia. There may be cultivated strong perennial winter wheat.The morphological characteristics of hybrid F₁ was in the middle of parents,such as plant tall, lush, perennial, and cold resistance. The character of Hybrid F₂was segregated, and all hybrids are safe for the Harbin winter without covering.The preliminary judging for the morphological detection of hybrids showed that all progenies of hybrid had the genetic materials from Thinopyrum. Through the count of root tip chromosomes and the observation of meiotic, we made the judgments of the stability and chromosome configuration for progeny. Then, we used RAPD to detect all progenies, in which contained the DNA sequence of Thinopyrum intermedium. Finally, we identified the whole chromosome of Thinopyrum intermedium in progenies of hybrid by genomic in situ hybridization, and preliminarily estimated the stability of hybrid F₃, seed set, and perennial habit etc.
