Camellia oleifera is an endemic woody edible oil plant in southern China, which is called one of the world's four major woody edible oil plant together with oil palm, olive, coconut. In this paper, tissue culture techniques of Camellia oleifera were studied, the main results are as follows:1,In the tissue culture of Camellia oleifera, July was the best explant collection period in a year, and compared with 3-year-old bud explants, new cut bud explants of Camellia with 1-year-old could effectively improve the induction rate, and reduced the pollution rate and browning rates.2,After pretreatment, rapid disinfection with 75% alcohol for 30s, and 6 min 0.1% Hgcl2 disinfection, stems with buds of Camellia oleifera were inoculated with MS+6-BA2.0 mg/L+NAA0.2 mg/L medium, the induction effect of buds and callus was best. There was significant difference in the induction rate of Camellia among the three clones('cen ruan 2','cen ruan 3'and'gui ruan 11'), and their induction rates were 57%,67%, and 33%, respectively. In addition, the best induction effect of callus proliferation appeared after treated with 6-BA 2.0 mg/L and NAA 0.2 mg/L hormone in the medium.3,The MS medium containing distilled water, sucrose concentration of 30 g/L, agar 0.51% and pH 5.8 provided the best culture conditions for Camellia oleifera tissue. After inoculation, the effect of tissue culture of Camellia oleifera was better when the tissue was treated with direct sunlight training(1400 lx light intensity and time for light 12 h/d) than treated with dark for 7 days.4,The best medium for camellia oleifera embryogenic callus was MS+2,4-D2.0 mg/L+KT1.0 mg/L, and the induction rate was 73%. The embryogenic callus transferred to MS+6-BA0.6 mg/L+NAA 2.0 mg/L medium for embryoid induction had the highest induction rate (73%).5,The best medium for aseptic Seedlings of camellia oleifera is SH+6-BA2.0 mg/L+NAA0.2 mg/L.
