Tung tree (Vernicia fordii (Hemsl.)) is China's unique economic trees, as important industrial oil. And flower bud differentiation is the basis of the reproductive process of plants, directly related to fruit production and quality. In this paper,it studies response to the signal of hormone and, the infection of the diurnal variations of photosynthesis on vernicia fordii(Hemsl.) dealing with different exogenous hormones.The result of this study provides the theoretical basis for using the exogenous hormones.This study's major findings are as follows:1.Comparing the quality of total RNA that using different concentration of CTAB,different extacted method,defferent precipitated method and different dosage of sample,this paper presents the method of extacting total RNA.RNA detected by electrophoresis figure that 28S,18S,5.8S are clear,28S:18S is close to 2:1,OD260/OD280 is between 1.7 and 1.9.It is clear that the total RNA is separated using improved method of CTAB have good integrity and quality.2.Analyzing of Plantβ-Actin conservative gene sequences, design primers to fordii flower total RNA reverse transcription of first strand cDNA as a template,the. cDNA fragment of about 400bp by using RT-PCR amplification was cloned into pMD-18 vector.Sequencing and sequence analysis showed that the cDNA fragment of 396bp was obtained,and has a higher degree of nucleotide homology with Ricinus communis, reached 97%.3.Using fordiiβ-Actin as internal parameters.Using extraction of total RNA of the buds in different stages of flower bud differentiation period, the first strand cDNA of reverse transcription as a template of fordii that are treated by different exogenous hormones.Respectively, using theβ-actin gene primers and Tunglfy primers for RT-PCR amplification, gel electrophoresis with PCR products show that the ABA 100mg/L,to advance expression of Tunglfy, three different consistence 6-BA to restrain expression of Tunglfy.4.Choosing the fordii are treated by GA 200mg/L,100mg/L;ABA 200mg/L,50 mg/L;6-BA 100 mg/L,25 mg/L,using the Li-6400 system to determine the Pn,Tr etc.The result show that GA to advance the photosynthesis.The higher concentration of GA have more promoting effect on photosynthesis of fordii than lower concentration.
