Tung tree (Vernicia fordii (Hemsl.)) is China's unique economic trees, as important industrial oil. And flower bud differentiation is the basis of the reproductive process of plants, directly related to fruit production and quality. Many studies have been currently carried out on fordii, but the mechanism of its flower bud differentiation reported less. In this paper, on the basis of studies of fordii's flower bud morphological differentiation by microstructure observation, it has been studied of Tunglfy gene expression and the dynamic changes of hormones in leaves in different organs and Stages of its flower bud differentiation, with a view to an important physiological factor to explore the endogenous hormone changes in the relationship between flowering,and provided a theoretical basis for chemical regulation for the study of fordii's flowering the mechanism of the flower bud differentiation. This study's major findings are as follows:1.Analyzing of Plant Calmodulin conservative gene sequences, design primers to fordii flower total RNA reverse transcription of first strand cDNA as a template, the cDNA fragment of about 300bp by using RT-PCR amplification was cloned into vector PCR2.1 vector. Sequencing and sequence analysis showed that the cDNA fragment of 293bp was obtained, named VeCam,, and has a higher degree of nucleotide homology with Kiwifruit, reached 96%.2. Analyzing of gene Tunglfy fordii 3'end sequences, design the specific primers. After extraction of total RNA of the buds, leaves, stem in different stages of flower bud differentiation period, the first strand cDNA of reverse transcription as a template, respectively, with the VeCam gene primers and primers for RT-PCR amplification, gel electrophoresis with PCR products show Tunglfy gene expressied in bud leaves, stems and different stages of fordii's flower bud differentiation,relatively strong in bud.3. The changes of Endogenous Hormones during the Flower Bud Differentiation of Vernicia fordii(Hemsl.)Airy Shaw were studied. The results showed that the contents of ZR, GA, IAA and ABA decreased before the Flower Bud Differentiation,while the ratios of ZR/GA,ZR/IAA,ABA/GA,ABA/IAA increased;During the Flower Bud Differentiation,both of that the contents and ratios changed between the relatively fixed range in a higher level.From the end of Flower Bud Differentiation, the contents of ZR, GA, IAA and ABA increased slightly before the Flower Bud Differentiation,while the ratios of ZR/GA, ZR/IAA,ABA/GA,ABA/IAA increased;the balance and combined effects of endogenous hormones may be more meaningful to the flower bud differentiation and Flowering regulation.
