Currently, seaweed, growth regulator as the commercialized products has been promoted for agriculture in many countries. It has got the astonishing results,for examples, significantly improving the yield of crops, promoting plant growthand improving the resistance. Most scholars believe that it is duing to the bioactive substances such as betaine, alginic acid, fucosan and endogenous hormones.Seaweed growth hormones in China seaweed fertilizer are widely used day by day, thus, qualitative and quantitative analysis of these active materials in seaweed fertilizer are important. Nowadys how to reserve those active substances in production is the bottleneck for this industry.In this paper include four parts:Firstly, the method of determination of kinds of endogenous hormones in fresh kelp and Enteromorpha by High Performance Liquid Chromatography-tandem Mass Spectrometry (HPLC-MS/MS) has been developed. Extraction was first extracted from fresh kelp by 90% menthanol solution with ultrasonic, then purified by MAX and MCX solid-phase extraction column, and analyzed by the HPLC-MS/MS. As the results shown that recoveries of the method were in the range of 74.51%-96.21% for the kelp, and 73.97%-97.23% for the Enteromorpha. The detection limit of ZT and ABA were 4μg/kg·FW, and the quantitation limit were 10μg/kg·FW, the detection limit of IAA and IBA were 5μg/kg·FW, and the quantitation limit were 15μg/kg·FW. Samples from 5 different areas of Qingdao sea approach in July 2008 were determination by HPLC-MS/MS, as the results revealed that the content of IAA and IBA is higher than the content of ZT, and ABA is the lowest or no detection.The content of endogenous hormones in Enteromorpha from trestle area was significantly higher than the other four areas, IAA and IBA even reached to 194.21μg/kg·FW and 187.28μg/kg·FW. In the second part, the evolution of endogenous hormones from different parts and growth stage (young period, thick period and mature period) of Rongfu kelp was system analyzed. As the results shown that the four kinds of endogenous hormones in different parts were changing in the range as follows:ZT 8.06-49.77μg/kg·FW, IAA15.35-229.46μg/kg·FW, IBA18.75-109.72μg/kg·FW, ABA23.14-95.89μg/kg·FW.In the third part, the acid hydrolysis processes of the materials in Rongfu kelp were optimizing. In order to reserve the active substances, the twice-dipping method was used in the processing. With the orthogonal test, the best conditions for acid hydrolysis was attained, that the kelp should be acid hydrolysis with 3% acid A in 100℃for 6h.The content of alginic acid in the extract reached 19.32%, and content of endogenous hormones in seaweed fertilizer powder, which was determination with LC-MS/MS, was as follows: ZT 70.66ug/kg·DW, IAA 496.60μg/kg·DW, ABA 105.22μg/kg·DW, IBA 103.54μg/kg·DW. It was interests that the samples which were not produced by the twice-dipping method had lower contents, as follows:ZT 38.34μg/kg·DW, IAA 180.32μg/kg·DW, ABA 39.75μg/kg·DW, IBA 30.66μg/kg·DW. It was obviously that this new processing can improve the extraction efficiency significantly.Finally, the fertilizer efficiency of seaweed fertilizer produced by this new method above was proved. The experiments included indoor test and field cultivation. The liquid fertilizer, powder fertilizer were used as the contrast, then the four seasons cabbages were planted by the method of soaking and spraying. Indoor test shown that the liquid fertilizer and homemade seaweed fertilizer can increase the germination about 10%. After 20 days the plants spraying with liquid fertilizer and seaweed fertilizer were more stronger than the blank, the main stem increase about 0.2-0.3cm, the leaf areas were separately improved 89.3% and 80.5%, and the fresh weight increased 33.6% and 44.8%. The field experiment shown that homemade seaweed fertilizer can obviously promote the germination of the seeding, which increased the growth of roots, enhanced resistance to the lodging and increased the yield about 131.5%contrast to the blank.
