Promoting the improvement about long fiber cotton in molecule breeding to is very important for studying the forming about the tissue of G.barbadense in the paper.There wore the configuration changes and molecule mechanism in the development about G.hirsutum,but a little in G.barbadens.The embryonic callus (EC) of Xinhai 16 was used in this study.the prophase, and the cells in different periods,such as the EC changed into somatic embryo, the early somatic embryo and so on, were compared with the asepsis lamina by abserving the paraffin section a nd DNA .It concluded apoptosis happened in the forming of somatic embryo.The method that DDRT-PCR was used to find the different genes of prophase EC,shoud be the base for the improvement about long fiber cotton in molecule breeding and cytology.The main conclusions were as follow:1 The prophase, anaphase, early somatic embryo of EC and the asepsis lamina were compared by abserving the paraffin section.The result showed that the cell wall in early somatic embryo and the the asepsis lamina had the similar thickness.while the cells of the earlier EC were bigger,and in anaphase it came into the cell crops.2 The DNAs of the prophase,the anaphase,the early somatic embryo of EC and the asepsis lamina were compared.The results showed that the strip of the DNA was the shape of ladder,during the EC changing into somatic embryo.it approved that The prophase,the anaphase,the early somatic embryo of EC and the asepsis lamina were compared in this period.3 Twelve reproducible bands were gained by using the RNAs of the EC and the hypocotyl of asepsis seedling to DDRT-PCR.These bands were linked with pMDTM18-T,then transformed into DH5a.4 different positive fragments of the earlier EC were gained by selecting and sequencing.4 Four different positive fragments were compared in GeneBank.It showed that the fragments had not high homology with the similar genes.But the homology were high in the 19th and the gene of cornstarch synthase,the 38th and GMPase in tomato and the 55th and calmodulin-binding protein in arabidopsis thaliana.Though Blastx,the 38th has high homology with the genes of unkowned protein in grape and arabidopsis thaliana.
