Cotton is one of the most crops in the world and is also the important raw material of various clothes and furniture stuff. However, but production of cotton is faced with a variety of biological and abiotic stress. Verticillium dahlia is regarded as cancer of cotton and abiotic stress, like high salinity, arid etc, can also hinder the development of cotton.NAC transcript factor, one of the largest terrestrial plants special transcript factor families, plays a significant regulate role in the progress of gene expression about the response to biotic and abiotic stress.In the preliminary study of transcriptome sequencing about the analysis of sea-island cotton and upland cotton in response to Verticillium dahliae, a differential expression unigene fragment was obtained, called NAC. The open reading frame was amplified in this artic and was named GbNAC1-1, and this gene was belonged to the TERN category and contained the typical C, D, E substructure by evolutionary tree and homologous sequence alignment. According to subcellular localization, GbNAC1-1 was localized to the nucleus. In the experiment of spatial expression, GbNAC1-1 had the expression on root, stem and leaf, but it mainly concentrated on vascular bundle. By the analysis of QRT-PCR about cotton infected Verticillium dahliae, the expression of GbNAC1-1 was down-regulation within 72 hours after infected indicating that GbNAC1-1 played a positive role in defending Verticillium dahliae. The silence of GbNAC1-1 caused cotton disease severly with VIGS. Overexpression transgenic plants of GbNAC1-1 showed the growth and development of transgenic plants was better comparing with wild type plants, suggesting that GbNAC1-1 might be involved into the plant growth and development. The over-expression plants was infected by Verticillium dahlia, and the result showed the infection situation is more serious in WT. Under various abiotic stress(ABA, Mannitol and Nacl), the resistance of transgenic plants reduced.The research of this study made us have a primary comprehension of the function of GbNAC1-1, and the result laid the foundation of clarifying the function of NAC gene.
