Screening The Active Components In Traditional Chinese Medicine For Anti-Newcastle Disease Virus

This study explores the active ingredients from traditional Chinese medicine for anti-Newcastle disease virus, to develop the new, effective and environmentally antiviral medicine. Test methods: Collected some Chinese treatment medicine for anti-viral diseases from some prescription medicine, then selected 4 kinds of high frequency anti-viral herbs (Radix Astragali, Radix Scutellariae, Rhizoma Coptidis,Folium Isatidis) as the test material. The herb was crushed and extracted by ethyl acetate and ethanol as the alcohol extract. The alcohol extract were concentrated by rotary evaporation method and separated by 200-300 mesh gel silica chromatography and collected the eluting components. The 20 different polar components were concentrated and dried again. The anti-viral drugs were screened from these samples.Marc145 cells were used as a screening model by inoculating Newcastle disease virus(NDV). Four groups were established in this experiment. Prevention group: Marc145 cells were cultured for 20 h in the culture plate, and then added the different polar components in the medium, inoculated with the NDV after 8 hours cultured. The cytopathic effec(tCPE)was observed and the OD values were determined by MTT method 22 hours post inoculation. Treatment group: Marc145 cells were infected by NDV after cultured 28h,added samples 8 h post inoculation, cultured cells for 14 hours, and detected the OD values. Self-healing group: The cells were inoculated NDV but cultured in the normal maintenance medium. Control group: The cells were just cultured in the normal medium.Results:(1)The 80 compositions were separated from 4 kinds of Chinese herbal medicine by gel silica chromatography.. (2) Establish a cell model for screening anti-virus medicine: Marc145 cells were infected by NDV. The CPE was observed cells were observed and the OD values were determined by MTT method 22 hours post inoculation.(3) Find some active ingredients for Anti-NDV:Radix Astragali : There are 4 kinds of components have the effect of inhibitory NDV. The OD values of the four components were significantly higher than the Self-healing group (P <0.01). Five kinds of components have anti-viral effect in Treatment group, OD value of the five ingredients were significantly higher than Self-healing group (P <0.01.).Radix Scutellariae: There are 5 kinds of anti-viral active ingredients in Skullcap prevention group. The OD value of the 16th ingredients was significantly higher than the Self-healing group (P <0.05), the others were significantly different with Self-healing group (P <0.01). There are 6 kinds of anti-viral active ingredients in Skullcap treatment group . The OD value of the 7th ingredients was significantly different with Self-healing group (P <0.05), others were significantly higher than that of Self-healing group (P <0.01).Rhizoma Coptidis: Four kinds of components have anti-viral effect in Coptis prevention group, the OD values of ingredients were significantly different with Self-healing group (P <0.01). Three kinds of components have anti-viral effect in Coptis Treatment group, the OD values of the three ingredients were significantly higher than Self-healing group (P <0.01).Folium Isatidis: Three kinds of components have anti-viral effect in Folium prevention group, The OD value of the three ingredients were significantly different with Self-healing group (P <0.01). Four kinds of components have anti-viral effect in Folium treatment group, the OD values of the four ingredients were significantly higher than Self-healing group (P <0.01.).The results showed that the screening model established by Marc145 cell can effectively screen the antiviral active ingredients from Chinese medicine. There were some anti-viral components have been found from Radix Astragali, Radix Scutellariae, Rhizoma Coptidis,Folium Isatidis using this screening model.
