Follicle stimulating hormone receptor (FSHR) and luteinizing hormone receptor (LH/CGR) are two members of the gonadotropin receptor (GTHR), they are mainly in the gonads, play an important roles on the gonadal development and in the formation of gametes. Related studies have shown that they are expressed not only gonadal tissues.but also others, including the uterus, oviducts, breast, placenta and so on, which is the specific function is uncertain. Experiments also show that FSH and FSHR knockout mice showed female infertility, LH and LH/CGR knockout male mice sterile. FSHR and LH/CGR can be seen that play an important roles in the reproductive function of animals. However, the studied expression of FSHR and LH/CGR in the female genital organs of buffalo is under relatively smallBuffalo is one of the China's valuable and rare species resources, due to resistance to crude feed, feed conversion rate, high nutritional value of dairy products. As the labor force in th south also widely raised. However, there is something inherent reproduction characteristics on the buffalo, such as low reproductive rate, poor performance of the population, including seasonal estrus, silent estrus, not obvious expression of estrus, long calving interval directly restricts the reasons for the development of buffalo industry. FSHR, LH/CGR some polymorphisms may be associated with multiple births of animal characters, and some early traits associated with reproductive disease, so some of FSHR and LH/CGR mutations may serve as breeding buffalo new molecular breeding tag.According to the study, the research object is adult female buffalo in Guangxi, the system of nucleic acid and protein levels of the gonadotropin receptor expression in the female buffalo reproductive organs, also studied the relationship between the FSHR, LH/CGR polymorphisms and reproductive performance of buffalo. In order to improve water buffalo reproduction and reproductive endocrinology of buffalo laid a theoretical foundation, the main results are follows: 1. The studies on expression of gonadotropins receptors mRNA in the reproduction tissues in buffaloAccording to the study, the different stages of estrous cycle in adult water buffalo, reproductive organs (ovaries, uterus, oviducts) for the study, one-step extraction total RNA of tissue samples using Trizol, while taking advantage of RT-PCR, quantitative PCR to study the the expression of FSHR, LH/CGR on reproductive organs in Buffalo. The results showed that the expression of FSHR and LH/CGR mRNA in reproductive organs (ovaries, uterus, fallopian tubes) in female buffalo, the content of FSHR and LH/CGR mRNA in the ovary during the follicular phase were significantly higher than the luteal phase of ovarian (P<0.05); FSHR mRNA in uterus during the follicular phase were significantly higher than luteal phase uterus (P<0.05), LH/CGR mRNA in uterus during the luteal phase were significantly higher than follicular phase (P <0.05); the content of FSHR mRNA in the oviduct during the follicular phase were higher than in the luteal phase oviduct (P>0.05), LH/CGR mRNA in oviduct during the follicular phase were lower than the luteal phase (P> 0.05), but theirs' no significant difference. That FSHR and LH/CGR play the role of buffalo in different periods with different estrous. FSHR can promote ovulation during folliclelar phase and LH/CGR plays an important role during luteal phase and the heat of formation.2. The studies on location and quantitative of gonadotropins receptors protein in the reproduction tissues in buffaloAccording to the study, using immunohistochemistry and Western blotting techniques to study the FSHR, LH/CGR in Buffalo, distribution FSHR, LH/CGR protein in different estrous cycle (folicular/luteal phase) buffalo ovaries, uterus and oviduct, and distributed in different parts of the reproductive organs, including ovarian granulosa cells, ovary membrane cells, luteal cells, endometrial epithelial cells, uterine gland cells and columnar epithelial tubal chorionic cells. Description FSHR, LH/CGR different target cells by ligand binding to participate in their buffalo reproductive control, and thus autocrine manner to regulate their physiological activities.3. The studies on gene polymorphism of FSHR, LH/CGR in buffalo.According to the study, the PCR-SSCP method was used in two genes FSHR exon 10 and LH/CGR exon 11 of polymorphisms in four varieties of buffalo. The results showed that in the local labor buffalo found two polymorphisms, the exon 10 of FSHR 984 T→C, the exon 11 of LH/CGR 221 G→C, but the individual mutation may be insufficient to explain the correlation with the reproductive performance of buffalo.In summary, this study demonstrated that FSHR, LH/CGR mRNA and protein are expression on the reproductive organs in buffalo, and as estrous cycle changes in the different dynamic; found two mutations that the exon 10 of FSHR and the exon 11 of LH/CGR. it is provide a theoretical basis for the buffalo reproductive physiology.
