Localization Of LHR And The Regularity Expression Of LHR MRNA In The Ovaries Of Estrous Cycle Phases In Fecundity Goat

The development and maturation of the ovum is a complex process,which works by many hormones, including gonad stimulating hormone,gonadal hormone,gonadorelin;gonadotrophin releasing hormone and so on.The LH is one of the most considerable hormone. As an excellent high accouchement local varieties, Jining gray goat expression of LHR is peculiarity in the ovaries of estrous cycle phases.And the peculiarity expression of LHR probably is one of the factors which result to Prolific.We research the localization and the gene expression of the LHR in the Ovarian by immunohistochemical and real-time fluorescence quantitative PCR. The experimental results are as follows:1.The expression of LHR mRNA in the ovaries of estrous cycle phases in Jining gray goat show the regularity,the expression of LHR mRNA in dioestrus is the lowest, the expression of LHR mRNA in protophase oestrus is begin step up, the expression of LHR mRNA in oestrus is the highest, the expression of LHR mRNA in anaphase dioestrus is begin to deduct rapid,and drop to the level of dioestrus.2.Through the results of gene expression analysis showed that, The ratio of LHR expression in the ovaries of estrous cycle phases is that:dioestrus: protophase oestrus: oestrus: anaphase dioestrus is (1.0±0.16):(15.73±2.59):(63.72±11.10):(2.04±0.63).The results showed that the expression of oestrus LHR is significant differences with other three period(p<0.01), the expression of protophase oestrus,anaphase dioestrus and dioestrus is quiet(P>0.05).3.The LHR positive material in the ovaries of estrous cycle phases are been seen in Theca cell,Granulose cell,megagametocyte and interstitial in blood vessel surrounding.The result shows that quantity of LHR-positive cells is increasing along with follicular developmental level in estrous cycle. The number of LHR-positive cell is the most in oestrus, The number of LHR-positive cell is the next in anaphase dioestrus, The number of LHR-positive cell is again next protophase oestrus,And the number LHR-positive cell is the least dioestrus.4.The ratio of the number of LHR positive cells as follows: dioestrus: protophase oestrus: oestrus: anaphase dioestrus is(35±4.04):(211±6.56):(60±4.58):(26±6.00), The number of positive cells is increasing with the follicular development and the increase of follicle size increased. the number of oestrus LHR is significant differences with other three period(p<0.05), the number of anaphase dioestrus is significant differences to dioestrus and protophase oestrus(p<0.05),And the number between protophase oestrus and dioestrus(P>0.05).
