Phalaenopsis is the most beautiful spiece of the Orchidaceae. Because it is loved by people all over countries, its scale of production is also growing. the long-term asexual reproduction and the external infection in recent years has caused a more serious problem of virus infection. This not only reduces the ornamental value of Phalaenopsis, making the leaves, flowers and buds form necrotic and lesions, but also causes huge economic losses to the Phalanopsis company. Therefore, virus elimination treatments and virus-free cultivation techniques to improve the ornamental value of Phalaenopsis has important economic value and practical significance.The most serious harmful viruses to Phalaenopsis are mainly CymMV and ORSV. Studies on the viru detection and removal have done on other types of Orchidaceae, but about Phalaenopsis these reports are so few. The thesis systematically studied the ELISA detection of CymMV and ORSV, and summarized the establishment of tissue culture system and the main methods of virus elimination. The main results are as follows:1. Established the tissue culture system of Phalaenopsis. The flower stalk buds as explants, the best surface sterilization is first disinfecting for 30s with 75% alcohol, and then sterilizing for 12min with 0.1% mercury; The best induction medium is MS+6-BA8.0mg/L+NAA0.1mg/L; The most appropriate multiplication medium is MS+6-BA5.0mg/L+NAA0.2mg/L; The best rooting medium is 1/2MS+6-BA2.0mg/ L+NAA0.5mg/L.2. Established the ELISA serological Detection method of CymMV and ORSV. The mature plants with diseases of Phalaenopsis'L008','1018'和'YSP'were detected by this method. Pedicels axillary buds of the mother plants have been induced. The plantlets were detected alse, and found the tissue culture process of the stalks can reduce the virus concentration appropriate.3. Systemly studied the meristem-tips culture of vitro plantlets, the larger meristem-tips culture of vitro plantlets, the chemical treatment with tips culture, the heat treatment with tips culture, and the combined with chemical and heat treatment of tips culture. Ribavirin concentration of 30mg/L, with temperature of 36℃, with tip length of 3±0.1mm is the best virus elimination method which has a higher survival rate and a better virus elimination results. In addition, the browning of the tips training process has been studied, and found that the activated carbon has the most obvious anti-browning effect, followed by the PVP. |
