The present study was conducted to evaluate the effectiveness of Ala-Gln and different feeding modes in growth, immune and anti-stress for CyprinusCarpio Var.Jian.The Ala-Gln was administered at concent rations 0.5% of dietary dry matter,which were fed to the Cyprinus Carpio Var.Jian of the density of 20 g/L in five different feeding modes:â… group(permanently control diet,control group);â…¡group(first 0.5% Ala-Gln-containing diet and then control diet at an interval of two weeks);â…¢group(before four weeks 0.5% Ala-Gln-containing diet and after four weeks control diet);â…£group (before four weeks control diet and after four weeks 0.5% Ala-Gln-containing diet);â…¤group (permanently Ala-Gln -containing diet). The experiment lasted for 8 weeks.Acute crowding stress was implemented at the last of experiment. Results indicated that:(1) Different feeding modes of Ala-Gln significantly influenced growth, feed intake and body composition in Cyprinus carpio var.jian(P<0.05). The growth ofâ… group were significantly lower than other four groups (P<0.05) which has significant difference (P<0.05). Weight gain rate and specific growth rate of theâ…¡group was significantly higher thanâ…¢,â…£,â…¤groups (P<0.05),â…¢andâ…£groups showed no significant difference (P>0.05).Average Weight gain rate and specific growth rate of theâ…¢group was significantly higher thanâ…¤groups (P<0.05), but protein efficiency were on the contrary (P<0.05).Viscerasomatic index of theâ…¡andâ…£groups was significantly lower than that of the control (P<0.05).Feeding rate of other four groups were significantly higher thanâ… group (P<0.05). Protein efficiency of theâ…¡andâ…¢groups were significantly higher thanâ… group (P<0.05), but feed coefficient were significantly lower thanâ… group andâ…¤groups (P<0.05). White muscle protein content ofâ…¤group higher thanâ… group (P<0.05) and others three groups showed no significant difference (P>0.05).(2) Different feeding schedule of Ala-Gln significantly influenced immunity and antioxidant (P<0.05). Serum SOD of theâ…£,â…¤groups were significantly higher thanâ… group (P<0.05) and theâ…¤group was significantly higher thanâ…¡,â…¢groups (P<0.05).GSH-Px in Hepatopancreas of other four groups which showed no significant difference (P>0.05)were significantly higher thanâ… group (P<0.05),But in serum,onlyâ…£,â…¤groups were significantly higher than that of the control group (P<0.05).LZM of the head kidney was more than spleen. LZM in the head kidney of adding Ala-Gln groups were higher thanâ… group,onlyâ…¡group showed no significant difference (P>0.05),LZM in the spleen ofâ…£,â…¤groups were significantly higher thanâ… group (P<0.05). NO in the head kidney of other four groups which showed no significant difference (P>0.05) were significantly lower thanâ… group (P<0.05)(3) Before acute crowding stress, HSP70 in serum of five groups showed significant difference (P<0.05),but the glucose and cortisol of serum on the contrary (P>0.05).The change trend of every group is similar at 0-72h after crowding stress. Serum cortisol achieved peak at Oh and glucose at 1h,then decline; HSP70 in serumre mained high level before 12h and restored the normal level until 48h.The peak of glucose of other four groups were lower thanâ… group.Glucose content of adding Ala-Gln groups were significantly lower thanâ… group after stressed 12h (P<0.05); Glucose content ofâ…¡,â…¢groups were significantly lower thanâ… group after stressed 48h (P<0.05).Every group had returned to normal level at 72h and not always significant (P<0.05). Theâ…¡group was the smallest of glucose rising rapidly after stress; glucose level ofâ…¡,â…¢groups were fastest return to normal state.The peak of serum cortisol of adding Ala-Gln groups were significantly lower thanâ… group (P<0.05).Serum cortisol of other four groups were significantly lower thanâ… group after stressed 1h and 3h (P<0.05);â…¡,â…¢groups were significantly lower thanâ… group in sixth hours (P<0.05);â…¢group was significantly lower thanâ… group in twelfth hours (P<0.05). There were no significant differences all groups after stressed 24h (P>0.05). Every group had returned to normal level until 72h and no significant difference (P>0.05).Serum HSP70 ofâ…£,â…¤groups were significantly higher than other groups(P<0.05)before acute crowding stress (P<0.05).Serum HSP70 ofâ… group was significantly lower than other four groups (P<0.05) after stressed lh,3h,6h and 12h;Andâ…¡,â…¢,â…¤groups was significantly higher thanâ… group (P<0.05), but not significant compared with IVgroup after stressed 24h. Until 48h or 72h every group had returned to normal level.And Serum HSP70 ofâ…£,â…¤groups were significantly higher than control group.In the present study it concludes that:adding Ala-Gln can promote the growth,immunity and Resistance crowding stress of Cyprinus carpio var.jian. There were significant differences of every feeding modes. Given economical efficiency and practicability suggested that intervals Feeding modes were recommended for the culture of Cyprinus CarpioVar.Jian.(4) The change trend of control group was similar in lymphocytes of spleen, head kidney and blood in vitro immunosuppression experiments. With the Ala-Gin level rising, the transformation of lymphocytes of head kidney was significantly improved,then significantly declined at 10mmol/L (P<0.05),but in the blood showed no significantly different at 10mmol/L (P>0.05), The transformation of lymphocyte of spleen have no significantly change (P>0.05). The change trend of immunosuppressive group was same. The transformation of lymphocytes of head kidney, spleen and blood was increase with the Ala-Gln level rising,but at 10mmol/L showed no significantly change (P>0.05). The influence of immunosuppression for kidney were more than spleen and blood.The change trend of control group was similar in lymphocytes of spleen, head kidney and blood in vitro restraint stress. With the Ala-Gln level rising, the transformation of lymphocytes of head kidney was significantly improved,then significantly declined at 10mmol/L (P<0.05) but in the blood and spleen showed no significantly different at 10mmol/L (P>0.05). The change trend of restraint stress group was same. The transformation of lymphocytes of head kidney, spleen and blood was increase with the Ala-Gln level rising,but at 10mmol/L showed no significantly change (P>0.05). The influence of stress for kidney were more than spleen and blood.
