In this study, the extraction, purification, modification, purity, physicochemical property, and antitumor ;tivity of Beet pulp were investigated.The main result are as follows:Polysaccharides are extracted by the methods of water extracting, precipitation with alcohol, and the optimum extract technology is determined by single factor and orthogonal experiments. The optimum parameters to ctract are as follows:the ratio of water to material 1:90, temperature 80℃, extracting time 2h, and extracting mes 2.In this condition, the extraction percent of polysaccharide is 3.74%; the best condition that the extraction f alcohol separated:Ethanol eventually concentration 80%,4℃,6 hours.the content of polysaccharide in beet by ethod of phenol-sulfuric acid is easy and quickly.Three methods are used to remove protein from BPS.The results show that the method of papain and TCA is etter than others.By using 0.1% protease, the polysaccharide solution is enzymic hydrolysis for 1.5hours in 40℃ater and pH4.0,centrifuge for removing protein, then add TCA until terminal concentration of 2%.In this (?)ndition, the rate of lost protein is 73.72%, and the polysacchrides contain rate is 90.29%.Consider chemical methods to decolour can make polysacchrides degradation, structural change. So use the tethod of adsorption. Active carbon can use its own adsorption and decolorization capacity to separate the color id polysaccharide. The optimum conditions were as follows:the decoloration temperature 60℃;pH3.0; the (?)sage of active cabon,0.4%; the decoloration duration,30min; the rate of discoloration reach 74.5%, the (?)lysacchrides contain rate is 9.67%.BPS without protein is separated into five fractions on cellulose DE-52 column chromatography which eluted ith of water and NaCl solution was used to separate fine polysaccharides. The major component BPS-D is eparated into two fractions on sephadexG-200 column chromatography, the bigger fraction BPS-D-G was lentified as a homogeneous component by methods of Sephadex G-200, HPLC and freeze-thaw test.The molecular weight of the Polysaccharide were measured by HPLC and sepharose CL-6B column hromatography,the average of BPS-D-G is 130kd,total sugar content is 95.7%.The unit monosaccharide of PS-D-G was identified as Galacturonic acid,arabinose,Galactose,Glucuronic acid, and glucose,the content is 5.9%,23.8%,13.0%,6.2%,1.1%.Infrared spectrum showed that BPS-D-G wereβ-glucoside bond.The specific (?)tatory of BPS-D-G is -16.In order to study the antitumor activities of different molecular weight of BPS, BPS-D, BPS-D-G,the in vitro rowth-inhibition of HeLa was evaluated by MTT tetrazolium assay, the concentration were 400ug/ml,800ug/ml, OOug/ml, 1000ug/ml,1200ug/ml, the results indicated that the different of BPS have antitumor activities in vitro, inally, the results showed that the highest inhibition activity of HeLa is BPS-D at the dosage of 1000ug/ml,...
