| The paper studied the rapid determination of fenvalerate residues by colloidal gold-based test strips. After optimizing the various factors and conditions of the test, fenvalerate residues colloidal gold-labeled strips were developed for the rapid on- site detection of fenvalerate residues. The fenvalerate competed with conjugate fenvalerate-BSA acting the coating antigen was immobilized on the nitrocellulose membrane were combine to artificial synthesized McAb, which was conjugated to colloidal gold.Gold chioride was reduced with sodium citrate to make 10nm colloidal gold, which was coupled with the McAb as immuno-colloidal gold. The optimum pH was 6.0.22μg of the antibody were needed to stabilize 1mL gold sol.The Millipore135 nitrocellulose(NC) membrane were selected as the materials. The conjugate fenvalerate-BSA acting as the coating antigen and rabbit anti-mouse IgG were immobilized on the nitrocellulose membrane as the test line (T line) and the control line (C line) separately. The sensitivity of fenvalerate was 250ng/mL and the detection time was about 10min.The experiment studied the interference with the detection of fenvalerate residues caused by the struetur alanalogue compounds.The result showed that all of the cypermethrin,tetramethri,bifenthrn,decamethrim,permethrin were not crossing reaction. The stability test indicated that there liability of the strip antigen was good at 4℃with in 24 months.The spiked and recovery experiments of rapeseed,flowering chinese cabbage,pakchoi,green pepper, samples were conducted with the methanol as the extract solvent, The conformation of this study was validated by HPGC. The average recovery of this method was 84%-98% and the coefficients of variation were under 10%.The results indicated that the test strips could be applied in the rapid detection of fenvalerate residues.The diagnostic strip detection was sensitive, specific and non-instrument, which could be finished within 15 minutes and the could be visually looked by eyes without instrumentation. The stability and simplicity make it effective to determine the large number of fenvalerate. It can be suitable for fast fenvalerate test in agricultural. Besides, the method could be used as reference for the detection of other veterinary drugs residues. |
