| This study examined the effects of aspergillus oryzae and saccharomyces cerevisiae on rumen fermentation and blood bio-chemical parameters in dairy cow. Firstly,I screened the sole adding dose and their appropriate proportion and mixed adding dose of aspergillus oryzae and saccharomyces cerevisiae in vitro.Then Chinese Holstein cows fitted permanent rumen fistula were chosen to study the results of the experiment in vitro by applying 3×3 Latin square trial. This paper was aimed at discussing Isoacids's regulation on rumen and providing references for improving digestion and productivity of dairy cow from the results of effective degradation rate of nutrient of diet in rumen, apparent digestibility of dairy cow, internal environment of rumen, production of rumen fermentation and cellulase activity.1.Screening the sole adding dose and their appropriate proportion and mixed adding dose of aspergillus oryzae and saccharomyces cerevisiae in vitro.(1)Add a separate trial of aspergillus oryzae and saccharomyces cerevisiae controlled trial design with single factor,the level of aspergillus oryzae added 0,0.02%,0.04%,0.06%,0.08%,0.1%DM 6 gradient respectively; saccharomyces cerevisiae added 0,0.17%,0.33%,0.50%,0.67%,0.83%DM 6 gradient respectively.Every level with 3 repeats and 0h,6h,12h and 24h time points in vitro.Composite index using a number of indicators on the medium pH,ammonia concentra,liquid residues in vitro DM,NDF and ADF disappearance rate of a comprehensive assessment,screening the suitability of aspergillus oryzae and saccharomyces cerevisiae were added, aspergillus oryzae was 0.04%DM and 0.08%DM, saccharomyces cerevisiae was 0.33%DM and 0.67%DM.(2)Mixing ratio of aspergillus oryzae and saccharomyces cerevisiae selection using L4(2~3) orthogonal design.The initial screening is aspergillus oryzae and saccharomyces cerevisiae to 4 groups of orthogonal and then cultured in vitro.Selected ratio of aspergillus oryzae:saccharomyces cerevisiae is 4:67.(3)Add levels of mixed of aspergillus oryzae and saccharomyces cerevisiae is as the same as(1).The adding dose were 0,0.15%,0.3%,0.45%,0.6%,0.75% DM 6 gradients.The appropriate proportion of mixture was 0.3%DM and 0.6%DM.2.The effects of aspergillus oryzae and saccharomyces cerevisiae on the dairy cow of rumen fermentation and the blood parameters.The feeding experiment of diary cow was designed by 3×3 Latin square trial. The adding dose of aspergillus oryzae was designed as 0,0.04% and 0.08% DM. The adding dose of saccharomyces cerevisiae was designed as 0,0.33% DM and 0.67% DM.The mixed of aspergillus oryzae and saccharomyces cerevisiae were 0,0.3% and 0.6% DM. In this part, the effects of aspergillus oryzae and saccharomyces cerevisiae on effective degradation rate of nutrient of diet in rumen, apparent digestibility of dairy cow, internal environment of rumen, production of rumen fermentation and cellulose activity were studied.(1)There were no significant difference in rumen pH among treatments which were levels of sole and mixed aspergillus oryzae and saccharomyces cerevisiae.The sole levels of aspergillus oryzae and saccharomyces cerevisiae decreased the concentration of ammonia nitrogen,but there were no significant difference(P>0.05), 0.3% DM mixture of aspergillus oryzae and saccharomyces cerevisiae significantly decreased the concentration of ammonia nitrogen(P<0.05). Rumen liquid acetic acid,butyric acid,TVFA and C2/C3 of 0.04% DM aspergillus oryzae were significantly higher than the group without adding them (P<0.05); Rumen liquid acetic acid,TVFA and C2/C3 of adding saccharomyces cerevisiae were significantly higher than the group without adding them (P<0.05); Rumen liquid acetic acid and TVFA of adding mixture of aspergillus oryzae and saccharomyces cerevisiae were significantly higher than the group without adding them (P<0.05).(2) Sole and mixed adding aspergillus oryzae and saccharomyces cerevisiae significantly improved effective degradation rate of nutrient substance in diet (P<0.05). 0.04% DM of aspergillus oryzae significantly improved apparent digestibility of CP and NDF (P<0.05),and it also improved apparent digestibility of other nutrient substance. 0.33% DM of saccharomyces cerevisiae significantly improved apparent digestibility of CP(P<0.05),and it also improved apparent digestibility of other nutrient substance. 0.3% DM of mixture significantly improved apparent digestibility of CP(P<0.05), and it also improved apparent digestibility of ADF.Each level of mixture improved apparent digestibility of DM and NDF(P>0.05).(3)Sole and 0.3% DM mixed adding aspergillus oryzae and saccharomyces cerevisiae significantly improved cellulase activity respectively(P<0.05),and the effect were perfect when the diet was added 0.04%DM aspergillus oryzae and 0.33% DM saccharomyces cerevisiae.(4) Sole and mixed adding aspergillus oryzae and saccharomyces cerevisiae improved serum calcium,serum phosphorus and urea nitrogen of dairy cow(P>0.05). Adding aspergillus oryzae significantly improved alkaline phosphatase activity(P<0.05). Adding saccharomyces cerevisiae and their mixture had no significantly effect on alkaline phosphatase activity(P>0.05). Sole adding aspergillus oryzae and saccharomyces cerevisiae had no significantly effect on glucose concentration(P>0.05),and adding 0.3% DM their mixture significantly improved glucose concentration(P<0.05). Sole and mixed adding aspergillus oryzae and saccharomyces cerevisiae significantly improved total protein concentration(P<0.05). |
