Effects Of Aspergillus Oryzae Culture On Rumen Fermentation And Microbial Diversity | | Posted on:2016-08-29 | Degree:Master | Type:Thesis | | Country:China | Candidate:J Xiao | Full Text:PDF | | GTID:2283330479481803 | Subject:Animal Nutrition and Feed Science | | Abstract/Summary: | PDF Full Text Request | | The paper mainly studied the topic from two directions of in vivo and in situ-the effect of Aspergillus Oryzae culture on rumen fermentation and hay dynamic degradability of sheep by the nylon bag and nutrients infusion technique at the same time analyzed the pH,NH3-N,VFA and the microflora of sheep.Firstly,the diets were divided into four groups and added 0%DM, 0.2%DM, 0.4%DM and 0.6%DM Aspergillus Oryzae culture to get the suitable addition according to the rumen fermentation and fiber degradability of sheep.0.4%DM Aspergillus Oryzae culture significantly improved the concentration of acetic acid and TVFA(p<0.05). Supplementation with Aspergillus Oryzae culture improved the degradability of fiber after fermenting 12 hours in the rumen. And 0.4%DM Aspergillus Oryzae culture have better effect on fiber degradability than 0.2%DM and 0.6%DM Aspergillus Oryzae culture.Secondly, using the suitable addition of Aspergillus Oryzae culture from first part result to evaluated the differences between Aspergillus Oryzae culture and Amaferm according to the rumen kinetics parameters, fiber and nutrients flows to the alimentary tract of sheep. The results showed as follows:(1) The Amaferm cultures groups had the biggest rumen liquid volume and retention time, on the contrary, Aspergillus Oryzae culture groups had the biggest rumen digesta volume and retention time.(2) There was no significantly difference between the different group of the nutrients flows in the same digestive tract(P>0.05).But supplementation with Aspergillus Oryzae culture decreased the nutrients flows.(3) Although dietary supplementation of Aspergillus Oryzae culture had no significantly difference on DMã€OMã€NDF and ADF in the same part of digestive tract of sheep(P>0.05),they can increased the nutrients digestion.Finally, a lot of papers pointed out that Aspergillus Oryzae culture stimulated the rumen fermentation and the ruminant productivity. The using of Aspergillus Oryzae culture was just like a black box because there was few research about the function mechanism and the effect on microflora of rumen. The last part of this paper studied the effect of Aspergillus Oryzae culture on the microflora of rumen. The special bands of microflora samples were further eluted from gel, reamplified and sequenced carefully.
