In this paper, plant species from the SaiHanWuLa Natural protection center were collected and examined to research the mycorrhizal status of the roots and the species of AM fungi. The spore of AM fungi were gotted from soil trap culture. Through field sampling and laboratory culture to investigate the initial distribution and species diversity of AM fungi. To provide theoretical basis for the classification, physiology, metabolism and the production. The AM fungi can be used in Agricultural and forestry development and protection of the ecological environment. The main conclusion is as follows:1.24 species of wild herbaceous plants were collected in the sample areas of SaiHanWuLa Natural protection center, belonging to 15 families and 21 genera.Among of them,22 species Mycorrhizal plants are collected and accounted for 91.7%, And 2 species Artemisia gmelinii and wintergreen are not infected by AM fungi.2.Nineteen spcies of AM fungi were isolated from the soil of plant species of Saihanwula. Eight species belong to Acaulospora.Seven belong to Glomus. Three belong to Diversispora. One belong to Scutellospora. Four species were uncertain. They are Acau. sp1, Acau. sp2, G. sp1 and G. sp2.3.The AM fungi isolated from all soil samples on the pot trap culture, are 4 genera and 15 species, compared with the original soil samples decreased by 4 species, Acau. elegants, Acau. rehmii, Dive. spurcum, Acau. sp2. For Acau. leavis and Scut. calospora species, trap culture are the most appropriate training methods, the spore production is increased significantly. Dive. versiforme fungi is second, and for G. constrictum show obvious maladjustment, other types of AM fungi spore density is no significant difference before and after culture. With single spore culture 5 species were established, Acau. leavis, Scut. calospora, Dive. versiforme, G. mosseae and G. intraradices.4.Dive. versiforme, G. mosseae germination rate was high in the water agar, and compared with other substrates is significantly different. in sterile soil + Sorghum substrate, Dive. versiforme, G. mosseae, G. intraradices germination rates is higher than sterilized soil matrix, the results show that there are living under the conditions of host plants, AM fungi can get a high germination rate. Different AM fungi has different rate, G. mosseae after cultured 7 days in water agar, the germination rate is the largest, but G. intraradices, Dive. versiforme are different, after 14 days can reached a peak, the maximum germination rate is also different.
