By studying the effect of neutron radiation on plant regeneration, Single plant screening for powdery mildew resistance and somatic clone establishment in red clover and study on resistance physiology, the results were as followed.1. After different dosage of neutron radiation, induction rates of cotyledons and hypocotyl were lower than the control. The highest induction rate for cotyledons and hypocotyl was obtained with the combination of 2,4-D 2.0mg/L and 6-BA 0.5 mg/L. MS+0.06mg/L NAA+0.1mg/L KT+ 2.5% Sucrose+0.7% agar was the best differentiation medium. Callus with light yellow color, loose granular structure and no hairy roots were obtained while cultured under this medium. 1/2MS+0.05mg/L NAA+1.5% Sucrose +0.8% agar was best rooting medium for Minshan red clover.2. Neutron radiation decreased survival number and rate of cutting shoot and with a survival rate of 0.66 to 2.27%. Survival rate in summer is higher than in autumn. Neutron radiation had some influence on plant height of M1. With the increasing of radiation dosage, branching number decreased in the next year. Branching number of the treatments were lower than the control except for the dosage of 1.2995Gy. One creeping plant occurred among the cutting shoots.3. The highest SOD activity (223.73U/g.min), POD activity (382.95U/g.min), PAL activity (105.70U/g.min), soluble sugar (0.0318%), Pro (29.93μg/g), Chlorophyll a (1.563 mg/g) and b contents (0.522 mg/g) were obtained among immune plants and they were 2.75, 1.13, 1.32, 2.41, 2.06, 1.23 and 1.50 times higher than those high susceptible ones, respectively. With the decreasing of disease resistance, all indexes decreased. For carotenoid content, no significant differences existed among different palnts.
