This study selected Reaumuria soongorica as research material to explore the genetic diversity differences of the natural populations and within populations in Zhangye, Wuwei, Jiuquan, Gaolan, Lanzhou. Through research the genetic diversity of different population of Gansu province R. soongorica, the major conclusions were described as follows:1. The optimal ISSR reaction system and reaction conditions of Reaumuria soongorica were finally established: 20μL PCR reaction system included 20ng template DNA 2μL,1.0μmol·L-1 primer 2μL, 10×Taq Buffer 2μL, 2.5U/μL TaqDNA polymerase 1μL, ultrapure dNTPs 2μL, distilled water 11μL. Reaction conditions: 94℃3min, 94℃denaturation 30s, 50℃annealing 45s (annealing temperature varies with the primers), 72℃extension 90s, 40 cycles, final 72℃extension 5min.2. 69 loci was amplified in the study, of which 60 was polymorphic loci. And polymorphic rate was 86.96%; DNA molecular fragment weight was between 500-3000bp. The highest ratio of polymorphic loci was Jiuquan (JQ) population, reaching 81.16 %, the lowest were Gaolan (GL) and Renshou mountain (RS)which was (75.36%). The order of polymorphic rates of five populations was: Jiuquan (JQ )> Wuwei (WW)> Zhangye (ZY)> Gaolan (GL) and Renshou mountain (RS).3. Each species Shannon index (I) polymorphism was between 0.4701 to 0.5152, and the average level was 0.4893,and the highest population was Jiuquan which was 0.5152, and the lowest population was Gaolan which was 0.4701; Nei genetic diversity index polymorphism (H) of each species was between 0.3281 to 0.3617 and the average level was 0.3424, and the highest population was Jiuquan which was 0.3617, and the lowest population was Gaolan which was 0.3281. The results of Polymorphic loci (P), Shannon index (I) and Nei gene diversity index (H) were basically same. The order of the various groups was: Jiuquan (JQ)> Wuwei (WW)> Zhangye (ZY)> Renshou Mountain (RS)> Gaolan (GL).4. The genetic similarity coefficient and genetic distance of different populations of Gansu province were obtained by Popgen32 . There had a high degree genetic identity between the populations , which the average was 0.9308. Poputions of ZY and JQ had the highest similarity coefficient, which were 0.9632; Poputions of JQ and RS had the lowest similarity coefficient, which were 0.9064. The average genetic distance of the five populations was 0.0478. The genetic distance between JQ and ZY was the shortest, which was 0.0375. The genetic distance between JQ and RS was the farthest, which was 0.0982.Using the UPGMA soft clustered and mapped to the five populations according to the genetic distance between the populations, and the results of the clustering was divided into three groups: ZY, JQ and WW were considered to be the first group. GL and the first group were thought to be the second. The third group was composed whith the first group, the second group and RS population. Mantel test showed that the genetic distance and geographic distance hadn′t significantly correlated.
