Technique Of Virus Elimination And Industrializing Propagation Of Jixin Jujuba (Zozyphus Jujuba)

The main results are:â…  To eliminate the pathogen of witch-broom disease of Jixin jujuba , tissue culture is the first way to chose. 38℃ to the test-tube plantlet, constant temperature of 60 days is dealt with.â…¡ The best explants is the new buds from roots. The sterilization method of explants body surface is:70% alcohol for 30 s, 0.1% HgCl₂ for 5 min, then transfers the explants into the culture medium: MS+6-BA1.0+NAA0.2, the survival rate is the highest.â…¢ The suitable medium for test-tube plantlet is: MS+6-BA1.5+IBA0.5;The best medium for test-tube plantlet root culture is: 1/2 MS+IBA1.5+NAA0.1+IAA0.5.â…£ The leaves of Jixin jujube can be the materials of plant regeneration. It can be turned into reality through producing callus\root tissue and bud plantlet. The middle part of leaves is suitable, put the leaves to face upward , with 1/2 petiole ,the petiole is inserted lightly into the medium, the suitable medium is: MS+6-BA1.0+NAA0.4. It can directly regenerate bud plantlet;It can produce callus in medium: MS+6-BA2.0+IBA0.5.â…¤ It does not need to open the bottleneck for test-tube plantlet practice of Jixinzao . The suitable time will be the middle or the last ten days of April. It is necessary to regulate the temperature\humidity\illumination and chose the peat soil disinfected. Peat soil: Vermiculite : Perlite =1: 1: 1 or Peanut chaff : Thick sand :Vermiculite= 1:1:1 mix soil is suitable.VI The material plant of non-test tube rapid propagation should be planted in net work room. The time for transplant is June to August, the cutting should be dipped into the root liquid. NO.3 root liquid has the highest root rate.
