The major cultivars named No.2 of Jiulong , No.12 and No. 2 which from Guangxi Luocheng were used as experiment material .The shoot tips and burgeons were used for explant which can be directly induced tufty buds and carried through rapid propagation ,and then, rootless sdddlings were attained. The methods of heat treatment and tip culture were combined to induce virus-free buds.Through induction,differentiation, multiplication, rootage and transplant, It studied the problem of medium, hormone proportion, season of taking material, position of taking material , sugar, browning and vitrification in the tissue culture. It also stuied the situation of virus-free with molecule method and tested the Physiological index.The results showed that the season of taking material and position of taking material all affect the effect of antisepsis and the rate of sprouting. The best month of collecting explant was from Marchl to May in which can attain the higher rate of survial, less pollution and better sprouting. Taking the middle and top , especially the top,were propitious to gain higher rate of survival and less pollution.The Sucrose was better than the Dextrose to induce buds.1/2MS was better than MS in the same case.Buds induced well on 6-BA+IAA,but on 6-BA+NAA was worse, especially,on 6-BA2.0mg/mL+IAA0.2mg/mL,the buds growed best.On the multiplication culture,the culture medium appendind MS+6-BA1.0 mg/mL+IAA0.01mg/mL was beneficial to growing and the plants grew rapidly , above all, coefficient of reproducing got to 4.0.Explant quite to be easy to appear the brown phenomenon in the process of induction. This experiment used the activated charcoal comparing with PVP , and then discovered PVP was more idealer. The browning was suppressed to be best when density was 1%. In multiplication culture, for reducing the vitrification phenomenon, let them in the 3000LX to culture.Rootless plantlets were inoculated in the 1/2MS culture medium in which appended BA0.2mg/L +IBA0.6mg/L.
