Recently, Immunosuppression and stunt syndrome was a new infectious disease which caused severely jeopardize in chickens. A novel virus(ISSV) was isolated from broiler chickens with immunosuppression and stunt syndrome. In the study, biological behavior, physiochemical criteria and morphologic were identified. Indirect enzyme-linked immunosorbent assay (IELISA) and indirect immunofluorescence antibody test (IFAT) were established for detecting the virus.1. After 48h post-inoculation, virions were observed in allantoic fluid from artificial infection SPF chicken embryos with isolated virus by electron microscope with negative staining. The virion was 40~70nm in diameter, anomaly hexagon, appropriately spherical or ellipse, with 3~5nm protuberance. Similar virions were observed in the thin-sections of proventricular, small intestine, liver and kidney of affected SPF chicken embryos and chickens by electron microscope.2. The biological behavior experiment showed that the isolated virus was RNA nucliec type. The virus could be propagated by inoculation of embryonated chicken eggs by allantoic route. Embryos were caused of regularly death on 3day to 5 day post-inoculation. When the virus infected SPF 10-day-old embryonated eggs artificially, at 72 hour post-inoculation was the peak during virus propagation. The virus couldn't be propagated using Vero, PK15 and CEF cell cultures.3. Physicochemical criteria assays showed that the virus could resist 20% aether and 4.8% choroform. This indicated the virion was naked viruses. The virus could resist completely at pH4 to 10.0, but only small virus survived at pH3.0 and lost the multiplication capacity completely at pH11.0 at 37℃ for 2 hour treatment. At 60℃ for 30 minutes treatment, the virus lost the multiplication capacity completely, but majority virus could survive at 56℃.4. The hyperimmune rabbit serum of anti-ISSV was prepared.
