| Recently years,Chicken Stunt and Immunosuppression Syndrome(tentative name,abbreviation:CSIS) was an importance infectious disease that mainly causes the suppression of growth and immunity on poultry.Based on the prophase research, this text will research the effect of Stunt and Immunosuppression Syndrome Virus on CD3~+,CD4~+,CD8~+,SIgA,ND antibody and IBD antibody of broilers,in order to lay the foundation to further research the immunosuppression mechanism of CSISV,and prevent and cure this disease preferably.Five-days-old ROSS308 broilers was artificial infected with CSISV,The contents of CD3~+,CD4~+,CD8~+ T cells were checked by using flow cytometry with FITC/PE double-color-staining method,then calculate the ratio of CD4~+/CD8~+;The contents of SIgA in blood serum were detected by application of indirect enzyme-linked immunoassay(ELISA) kit;Hemagglution Inhibition(HI) experiment was used to detect the ND antibody in blood serum;Agar diffusion reaction was used to detect the IBD antibody in blood serum;The datas were expressed by mean±SD, the difference between test group and control group was analyzed by t test with SPSS11.0 software.(1) The weight of broilers'main immune organs are all significantly lower than control group within a certain period,but incompletion at equal pace.The thymic weight are significantly lower than control group from 168h to 408h after artificial infection;The splenic weight are significantly lower than control group from 336h to 456h after artificial infection;The weight of bursa are significantly lower than control group from 216h to 336h after artificial infection.(2) Compared with control group,the difference of CD3~+ level in blood is not significant,but the difference of CD3~+ level in main immune organs is significant in different time.The integer tendency of CD3~+ is decreasing on 24h,increasing before 72h,after artificial infection 96h all lower than control group.The CD3~+ level in blood are higher than control group from 24h to 192h;lower than control group from 216h to 432h,but both the differences are not significant(P>0.05).The level of CD3~+ in thymus are very significantly(P<0.01) lower than control group on 24h; significantly higher than control group(P<0.05) from 48h to 72h;lower than control group from 96h to 432h,but the difference is significant(P<0.05) only from 168h to 264h.The level of CD3~+ in spleen are lower than control group on 24h;higher than control group from 48h to 96h,but both the difference are not significant(P>0.05); lower than control group from 120h to 432h,but the difference is significant(P<0.05) only on the 120,192,216 and 288h.The level of CD3~+ in bursa are higher than control group from 24h to 48h,but the difference is not significant(P>0.05);lower than control group from 72h to 432h,and the difference is significant(P<0.05) on 72,168,216~264,384h after artificial infection.(3) The level of CD4~+ in blood and main immune organs are different from control group significantly in different time.The level of CD4~+ in blood are higher than control group on 24h,but the difference is not significant(P>0.05);lower than control group from 48h to 408h,and the difference is significant(P<0.05) on 48~96h and 144h after artificial infection.The level of CD4~+ in thymus are lower than control group from 24h to 360h,but the difference is significant(P<0.05) only on 144h and 216h;higher than control group from 384h to 432h,but the difference is not significant(P>0.05).The level of CD4~+ in spleen are significantly(P<0.05) lower than control group from 24h to 48h;higher than control group from 72h to 192h,and lower than control group from 216h to 432h,but both the differences are not significant(P>0.05).The level of CD4~+ in bursa are higher than control group from 24h to 168h,and the difference is significant(P<0.05) from 24h to 72h;lower than control group from 192h to 432h,but the difference is significant(P<0.05) only on 216h and 312h.(4) The tendency of CD8~+ in thymus,spleen and brusa are coincident,the tendency of CD8~+ in blood from 96~288h is coincident too.The level of CD8~+ in thymus are lower than control group on 24h,but the difference is not significant (P>0.05);higher than control group from 48h to 216h,and the difference is significant(P<0.05) or very significant(P<0.01) on 72,96,120,192 and 216h.The level of CD8~+ in spleen are higher than control group from 24h to 264h,and the difference is significant(P<0.05) on 48,72,240 and 264h.The level of CD8~+ in bursa are all higher than control group from 24h to 288h,and the difference is significant(P<0.05) or very significant(P<0.01) on 24,48,72,120,192,216 and 384h.The level of CD8~+ in blood are lower than control group from 24h to 72h,but the difference is not significant(P>0.05);higher than control group from 96h to 432h, but the difference is significant(P<0.05) only on 216h and 264h after artificial infection.(5) The ratio of CD4~+/CD8~+ in thymus,spleen,bursa and blood are nearly all lower than control group on the whole experimental stage.Compared with control group:The difference of CD4~+/CD8~+ in thymus is significant(P<0.05) from 24h to 192h;The difference of CD4~+/CD8~+ in spleen is significant(P<0.05) on 24~96h and 384h;The difference of CD4~+/CD8~+ in bursa is significant(P<0.05) on 24,48,168 and 240~288h;The difference of CD4~+/CD8~+ in blood is significant(P<0.05) on 144~240,336 and 360h(6) After artificial infection,CSISV promte the production of SIgA from 96h to 264h,but suppress the production of SIgA from 288h to the end of the test;In addition,CSISV suppress the production of ND antibody and IBD antibody obviously on the whole experimental stage.The contents of SIgA in serum of test group are higher than control group from 96h to 264h after artificial infection,and the difference is very signidficant(P<0.01) on 96,192 and 216h;lower than control group on the other time,and the difference is significant(P<0.05) on 24,48,72,312,336h.The ND antibody titers of test group are lower than control group obviously on the whole experimental stage,on 22-days-old,the ND antibody titers can decrease by 2.33 at most,and the difference is significant(P<0.05).The IBD antibody titers of test group are lower than control group obviously on the whole experimental stage,on 25-days-old,the ND antibody titers can decrease by 2.67 at most,and the difference is significant(P<0.05).In summary,on the metaphase and anaphase,CSISV has a certain depressant effect on cellular immunity and has an obvious depressant effect on humoral immunity of broilers. |
