In this study, the general application and polymorphism level of 754 pairs of micro satellite primers derived from P.trichocarpa genome were tested in the species of Pop ulrs' relative genera —Salix. And the primers selected were used in three primary applications —constructing a linkage map for S.suchowensis×S.erioclada, comparing it simply with two poplar linkage maps and analyzing the association between markers and traits. The results as follows:754 pairs of SSR primers from Populus were screened aganist one DNA sample Among them, 371 pairs showed ampliction, and the percent of general application was 49.20%.It indicated that SSR markers from Populus could used in Salix very well, and can provide a number of candidate SSR markers for the willow species genetics.To examine the transferability of SSR markers developed in willow, all available primers subjected to PCR for 2 parent samples and 6 hybrid samples. By polymorphism detection, 157 of 371 primer sets could reveal polymorphism among the hybrids, which accouted for 42.32% of primers seletected. Each SSR locus could show 4 alleles mostly. Different research had different intents , for example, if for the purpose of genetic diversity, it may show a higher polymorphism level.For mapping, 100 markers showing polymorphism were selected by screening aganist 94 hybrid samples from the F1 progeny of S.suchowensis and S.erioclada. Accounting by JoinMap 2.0, it constrcted a genetic linkage map which identified 9 linkage groups (36 SSR markers) with an average distance of 11.7 cM between markers. Linkage group included 2 to 13 markers and 4 markers on average.Comparing above 9 linkage groups with 20 linkage groups of P. deltoides × P euramericana genetic map and 19 linkage groups of P. trichocarpa×P deltoids map, there were 4 and 3 loci which were identified the same as in the two poplar linkage map. But the number of linkage group with each locus in three maps was almost different, and the reason might be that there were less markers distributed in this map.
