As a model species in tree genome research, Poplar has a good background in genomics andmolecular biology. In this paper, in order to better understand poplar genome, we obtained a F1population derived from Populus deltoids Bartr.cv.'Lux'(I-69/55)×P. simonii, the populationsize is more than600individuals.205individuals from the F1population were chosen asmapping population, a genetic linkage frame map of Populus deltoids Bartr.cv.'Lux'(I-69/55)×P.simonii was constructed based on SSR and SRAP markers. Meanwhile, the mappingefficiency was compared between two mapping softwares (Joinmap4.0and FsLinkageMap2.0),and the genetic maps of different poplar species were also compared. It would be useful in thefields as construction of high density genetic map, QTL mapping and marker assisted breeding inpoplars. The main results are as follows:(1)Construction of genetic map of Populus deltoids×P. simonii.With FsLinkageMap2.0,we acquired a genetic linkage frame map of108markers (88SSR markers and20SRAP markers)and30linkage groups. There were12linkage groups with4markers or more, and6thriplets and12doublets. The total map length was1462.52cM with an average of34.75cM, ranging from4.51cM to140.10cM. The average distance between adjacent markers was13.67cM, rangingfrom1.0cM to34.6cM.(2)The mapping efficiency of the two mapping softwares (JoinMap4.0&FsLinkageMap2.0). FsLinkageMap2.0is more powerful than Joinmap4.0in its closer adjacentmarkers, more markers, more linkage groups, and bigger map distance(3)Comparative maps for different poplar species. Based on54homologous SSR markers,2maps of Populus deltoids Bartr.cv'.Lux'(I-69/55)×P. simonii and P.trichocarpa×P.deltoideswere compared, and14homologous linkage groups and1presumptive homologous group werefound. According to information from29homologous SSR markers,2maps of Populus deltoidsBartr.cv.'Lux'(I-69/55)×P. simonii and P.deltoides×P.euramericana, were compared,7homologous linkage groups and5presumptive homologous groups were revealed. There wereonly7homologous markers between the map of Populus deltoids Bartr.cv.'Lux'(I-69/55)×P.simonii and the map of P.adenopoda×P.alba, and7presumptive homologous groups weredetected.
