Calla lily(Zantedeschia.spp) is a famous flower crop ,widely cultivated throughout the world. In recent years, with new varieties introduced into Gansu province, China, viral disease occurred in all these areas where calla lily was planted. And the influence of viral disease had been become more and more obvious. However, research work on viruses infecting calla lily remained scarcity in China. So, studying on the viral diseases of calla lily had greatly theoretical and actual significance. In this paper, viral pathogen naturally occurred on calla lily crops was identified systemically by biological and molecular techniques, which can lay a theoretical foundation for producing of calla lily virus-free seedlings, and provide technologies for detection and identification of viral disease of calla lily. The contents of this study mainly included investigation of the occurrence rate of viral disease, the study to forms of viruses particles and inclusions of cells infected with calla lily virus by electron microscope techniques, the extracting of plant total RNA, establishing of RT-PCR detection system of calla lily virus, the CP genes clone and homology analysis of sequence of calla lily viruses. The results showed that Zantedschia mosaic virus (ZaMV) was the viral pathogen infecting calla lily at natural condition in Gansu province, China. It was a species of the genes of potyvirus.Based on the survey of virus infecting calla lily in Gansu Province, the result showed that field incidence of ZaMV was 80.9%, 28.1% and 27.5%,respectively, in Jiayuguan city ,Yuzhong and Lintao county. Moreover, incidence of ZaMV of calla lily cultivated on the greenhouse was more than incidence of ZaMV of calla lily planted in the field.Natural-infected calla lily crops exhibited diversiform symptoms such as mosaic, mottling, malformation and so on. Host range tests suggested this virus had narrow host ranges, so it was very difficult to identify this virus by host reaction test. And ZaMV could only infect Araceae plants.Filamentous and flexuous virus particles, resembling potyvirus, were detected in 20 out of 38 samples by electron microscope techniques (EM).
