Zearalenone(ZEA), i.e.F-2 toxin,is an epiphyte toxin which has estrogen action produced by Gibberlla zeae, F. graminearum,Fudarium tricinctum and so on.It mainly empoison feed and cropper, such as wheat oat corn and so forth. It can arouse uterine portliness, metrohypertrophia, non-oestrus and abortion of female livestock, female symptom of boar. In addition,ZEA still has the immunity toxicity and latent cancer-causing, to the threat of the animal and mankind's health very huge.So the research of toxicity function caused by ZEA on the male animal germ cell has extremely vital significance, to enhances animal's reproduction performance and improves the livestock product quality.This research first carries on raise to Sertoli cells of the rat in vitro,then DNA damage of the big mouse testicle Sertoli cells in vitro cultured exposed to ZEA for 16h was determined by single-cell gel eleetrophoresis(SCGE), finally the effect of ZEA on expression of mRNA of androgen binding protein gene in vitro raised Sertoli cells exposed to ZEA for 24h was determined by one-step RT-PCR methods. Through these experiments we discuss toxic effect of ZEA to the male germ cell.The results showed that Sertoli cells which We obtain through in vitro raise accounted for above 90% in the culture cell total quantity. In SCGE experiment, ZEA had lead to the damage on DNA of Sertoli cells in the definite range of concentration. With the concentration increasing,the extent of damage also increased,which showed an evident logarithm dose-effect relationship.And except 5 μ g/mL group,the difference of the tail rate, average tail length and DNA damage in each group was unnoticeable compared with negative group(P<0.05) The ABP mRNA levels of Sertoli cells in vitro cultured were decreased with the increased of ZEA concentration,and there was a significant dose-effect relationship in RT-PCR experiment. Compared with control group , the decreases of ABP in each group was significant(P<0. 05).The conclusion showed that ZEA could cause DNA damage and inhibit the translation of ABP in the rat Sertoli cells cultured in vitro. Therefore ZEA has the toxic effect to the male reproductive system.
