Background:Zearalenone(ZEA)also called F-2 toxin,is a produced by Fusarium fungi with estrogenic activity and non steroidal mycotoxins,it has a special affinity with the estrogen receptor in vivo.Zearalenone is a kind of biological toxin with special toxicity,the toxic effect is mainly manifested in its strong estrogen like effect,Resulting in a decline in animal reproductive function,causing serious economic losses to animal husbandry.As one of the most effective natural antioxidants to scavenge free radicals in the human body,proanthocyanidins have a strong antioxidant activity,can effectively scavenge superoxide anion free radical and hydroxyl radical,and its source is very extensive,In this experiment,choose to use proanthocyanidins as antioxidants to study the effect of proanthocyanidins on the protection of zearalenone against oxidative damage induced by mice testis sertoli cells.Objective:The purpose of this experiment was to study in vitro the protective effect and mechanism of proanthocyanidins on zearalenone induced mice TM4 cell oxidative damage,the aim of this study was to provide theoretical basis for the prevention and treatment of zearalenone poisoning in animal husbandry production.Methods:MTT method was used to detect different concentrations of zearalenone and proanthocyanidins alone and in combination on mice TM4 cell relative survival rate;flow cytometry was used to detect the apoptosis rate of each group;detected the activity of T-SOD,LDH,GSH-Px and the content of MDA and GSH in each group by the kit;RT-PCR method was used to detect nuclear factor E2 related factor 2(Nrf2),heme oxygenase 1(HO-1),glutathione peroxidase(GSH-Px),quinone oxidoreductase 1(NQO1),?-glutamylcysteine synthetase(?-GCS)the expression level of Mrna;Western-blot method was used to detect the protein expression levels of Nrf2,GSH-Px,HO-1,-GCS and NQO1 in each group.Results:1.Zearalenone has a damage effect on TM4 cells,The low concentration of procyanidins have a promotive impact on the growth of cells,PC have protective effects on TM4 cells exposed to ZEA.2.Compared with the zearalenone group,procyanidins could reduce the apoptosis rate of the Sertoli cells exposed to zearalenone(p<0.01);Compared with the zearalenone group,Proanthocyanidins can enhance the activity of T-SOD and GSH-PX(p<0.05,p<0.05),reduce MDA content,significantly decreased LDH activity(p<0.05),significantly increased GSH content(p<0.05).3.Compared with the zearalenone group,proanthocyanidins can significantly up regulate the Nrf2 and GSH-Px,HO-1,-GCS and NQOl mRNA and protein expression levels.Conclusion:PC have protective effects on oxidative damage induced by ZEA in mice TM4 cells.Protective effect of procyanidins on zearalenone induced mice TM4 cell injury may be through the activation of the Nrf2/ARE pathway to achieve.
