In the past two decades, fish immunology has become a highlight of the fish preventive medical research, and has gained a lot of achievement in many areas. So far, most of the fish immunology researches were focused on fish primary immune system and peripheral immune system,only a few works had been done on mucosal immunity.Fishes live in the water,a "germ rich" environment.Fish has large mucosal tissues covered on them surfaces of skin ,gill and intestine, which serve as first barrier in the defense against disease. So, make further research on fish mucosal immunity would help us to comprehensively understand the pathogen invading, course of disease and recovery of disease. Fish mucus immunoglobulin (mIg) played an important role in preventing fish from disease, but little was known about fish mIg. By comparing fish mIg to fish serum immunoglobulin (Ig), figuring out their differences in structure and the antigenicity, illustrating the origin and place of the mIg, developing the methods for detecting fish Ig and mIg,monitoring fish immune level,evaluating fish immune response to the vaccination,it will contribute to further understanding of fish humoral immunity, and also will promote the development of fish immunology. In this dissertation, the purification of mIg and Ig of the European eel ,analyzing and comparing their structure and antigenivity were described.1.the affinity purification of mIg and Ig of the European eelThe purification of mIg and Ig of the European eel were carried out by ammonium sulfate precipitation and followed by affinity chromatography purification in which the monoclonal antibody(McAb) to the serum Ig of the European eel was used as ligand for catching the mIg and Ig. The results showed that after separating by the affinity column, the amount of eluted proteins of both mIg and serum Ig were showed a sharp curve when drawed the data on plotting paper .
