Objective: Porcine allergic action can be caused by injecting vaccines,especially some species. Allergic action can cause animal death without curing in time. Allergic action is mainly mediated by Ig E(Immunoglobulin E, IgE).Many methods are used in clinical,detect the content of serum IgE is high specificity and sensibitity and used widely,especially detect serm IgE by ELISA,the method is used in adult's allergic action, thus separation and purification of porcine serum IgE is the precondition and foundation of detecting porcine allergic action by ELISA. IgE ' s content is minimum in health adult, porcine serum IgE level hasn't accurate report.The purpose of this experiment is that through two methods extract IgE from blood serum of artificial infected cephalocyst and sensitized experimental swine,compare differences between the extraction of IgE from different serum samples and identify serum IgE by SDS-PAGE,thereby create the best method to extract IgE from porcine serum,so as to profound investgate separation and purification of porcine serum IgE and anti-IgE antibody and other serums by ELISA. Method:Artificial infected cephalocyst and sensitized experimental swine are two different samples,separate and extract IgE through aturated ammonium sulfatesalting out—SePhadex gel chromatography—DEAE ion exchange and salting out-ion exchange method-gel chromatography,and use SDS—PAGE electrophoresis detect and identify extraction IgE meanwhile,compare with porcine serum samples which also extracted and identified by the same method.Result: ?Extract IgE from blood serum of artificial infected cephalocyst and sensitized experimental swine by two methods,using aturated ammonium sulfate salting out—DEAE ion exchange -SePhadex gel chromatographygel ,and through SDS—PAGE electrophoresis show single band at 73KD; (2)using aturated ammonium sulfatesalting out — SePhadex gel chromatography—DEAE ion exchange to separate and extract the two samples,through SDS-PAGE,no IgE be extracted;(3)through the experiment IgE is not extracted from normal porcine serum exponents.
