According to the published nucleotide sequences of Duck Hepatitis B Virus from GenBank, two methods of PCR and fluorsescence quantitative polymerase chain reaction(FQ-PCR) for detection DHBV was developed in this study. They were used to the investigation of duck hepatitis B virus about molecular epidemiology and the establishment of disease-model of DHBV by artificially infecting ducklings. Disease-model of DHBV was used to select and evaluate the anti-HBV drugs.One pair of primers were designded and synthesized according to the consensus sequence of Duck Hepatitis B Virus from GenBank. Using the primers, a 215bp DNA fragment was amplified from DHBV samples, but it can not be amplified from Duck Plague Virus, Duck Hepatitis Virus, pathogenicity Escherichia coli, pathogenicity Salmonella, Riemerella anatipestifer samples, et al.The PCR method could detect 0. 42pg template. Using it to detecting Yaan Tianfu duckling, the natural carrying rate of DHBV was 12. 34% (38/326) .
