The maize genotypes of high inductivity of embryo callus and the factors of induce variation on maize young embryo in vitro culture was filtrated and discussed, at the same time, provided the theory support for the next research works in transgene breeding.The test with seven maize inbred lines 540, 7932, 7313, 5010, HQ2 and zong 31 as the material, carries on the young embryo culture in vitro, and analyses the influential factors on inductivity and rate of inducement of embryogenic callus (rate of EC)of maize young embryo callus through callus induction, secondary culture, differentiation and plant regeneration and regeneration plantlet rooting, hardening off, transplant and so on; optimize the method of maize young embryo culture in vitro and plant regeneration; carries on appraisal of regeneration plant on Cytology Botany Physiological biochemistry and Agronomic characters. The influence of genotype, young embryo size, 2.4-D and AgNO3 on callus inductivity was mainly analyzed in this test.The results indicated that only 540, 7313, 5010, HQ2 and zong 31 could induce embryogenic callus in the first culture, the rate of EC was 12.0%, 82.5%, 7.2%, 63.1%, 71.1% separately.
