Several aspects were researched about the tested strains of probiotics on the basis of the foundation of intestinal epithelial cells model of chick embryo: the adhering and anti-adhesion capacity of Lactobacillus Acidophilus and Lactobacillus bulgaricus; the change of Lactobacillus Acidophilus and Lactobacillus bulgaricus after treated by HCl-H2O, pepsin, heat, freese-thaw and so on; the anti-adhesion capacity of the metabolic products and lysate of the two tested strains of probiotics; the change of adhesion rate and anti-adhesion as the adhesion time changed; the influence on the anti-adhesion capacity by the adding time of the probiotics before or after the pathogenic bacteria; the inhibitory action on the pathogenic bacteria invading into the Intestinal Epithelial Cells model of Chick Embryo of the two tested probiotics.The foundation of the Intestinal Epithelial Cells model of Chick Embryo. The essay successfully applied I- collagenase and thermolysin to the isolation of lots of perfect pile-crypt- unit and the achievement ratio of cultivation added up to 85%. After 1~2 days, the pile-crypt- unit started adherence, and then began to"erupt"spindle cells. The spindle cells spreaded to the surrounding. After about 5days, they changed into cell population; After 10 days or so, they mixed together as a piece. The primary culture cells grew into monolayer under light microscope and the cells was thin and flat cerioid like shuttles and of clear boundary.The adhesion capacity of the two tested strains of probiotics. The adhesion rate of Lactobacillus Acidophilus and Lactobacillus bulgaricus not-treated was 21.00% and 8.73%; The adhesion rate of the two tested probiotics treated by HCl-H2O and pepsin decreased obviously; The adhesion rate of the twos treated by repeated freeze-thaw were not different from the control; The adhesion of the twos treated by heat were obviously higher than the control; The adhesion rate of Lactobacillus Acidophilus was significantly higher than the one of Lactobacillus bulgaricus; The adhesion rate stepped up gradually as the adhering time prolonged, but after 40 minutes the amplification of the adhesion rate decreased; 1~3×108cfu/ml was the ideal density which can make the probiotics achieve total adhesion.The anti-adhesion capacity of the two tested stains of probiotics. Lactobacillus Acidophilus and Lactobacillus bulgaricus not-treated had obvious anti-adhesion effect on Avian E. coli O78; The anti-adhesion capacity of the strains treated by HCl-pH2.0 and pepsin manifestly decreased; The anti-adhesion capacity of the ones treated by Buffer C had no obvious change; The metabolic products and lysate of the two tested strains had certain anti-adhesion effect on Avian E. coli O78;...
