Probiotics are live cultures of microbes,Which can altering the intestinal microbial balance to improve host healthy and growth. Probiotics have indeed shown to possess special health-promoting characteristics if they have the ability of colonization and adhesion.So,it's a hot to study the mechanism of adhesion.In the present,the in vitro screening model in order to bowel epithelioid macrophages Caco-2 lines, people colon cancer cell, various original HT-29 for cultivating the epithelial cells and purification mucous primarily. There is no report about normal immortalization neonatal swine intestinal epithelial cell lines in this filed.In the present research, Isolated the probiotics strains from piglets' excrement and different parts of fresh bowel.Took the Immortalization Neonatal Swine Intestinal Epithelial Cells ZYM-SIEC02 (abbreviation -n:SIEC)for the model , Discuss the adhesion, resistance and adhesion inhibition of pig Lactic acid bacteria. Designed to provide oretical basis for the mechanism of adhesion.In this stud- y,We obtained the following results:1.Got 12 Lactic acid bacteria strains. The research indicated that different strains have different adherence ability(p < 0.05) and culturefluid pH. Chosed the six high adhesion lactobacillus strains:the"Y09123"(2903.25±83.33CFU/100cells),"Y091221"(2320.37±81.02 CFU/100 cells)," Y224031"(1965.43±37.72CFU /100cells)had more stronger adhesion. In addition,the " X302032"(1416. 12±62.74 CFU /100cells), "Y223052"(1100.00±6.81 CFU /100cells)a -nd"Y223072("988.7 3±17.06 CFU /100cells)also have stonger adhesion. genera- lly,more stronger adhesion,more lower pH.2.Tested the tolerability and adherence abilitiy of the strains. Selected the better colones " Y091231"," Y091221"and" Y224031". Referenced the《Manual of identification in common clinical bacteria》and sequencing data of the conserved 16S rRNA genes showed that "Y091231" is Lactobacillus saliarius, "Y091221" is Lactobacillus johnsonii and"Y224031" is Enterococcus faecalis.3.Tested the antagonistic effect of Lactic acid bacteria against the adhesion of pathogenes. The result showed that all the 3 strains can reduce the pathogens' rate of adhesion obviously. Among those tests,the inhibition rate of" Y091231" against Escherichia coli C 83921 arrived (99.4±3.17)%, Similarly,the inhibition rate of "Y091221"against Enteritis salmonella arrived (97.1±2.04)% and the inhibition rate of" Y091231"against Staphylococcus CVCC2258 arrived (98.0±2.31)%. Those most high inhibition rates showed specificity antagonistic effect of different Lactic acid bacteria against the adhesion of diffrent Pathogenes.4.In the different growth periods,the strains have different adherence ability.when we cultered the testing strains to stationary phase(16h~20h),the adhesion number increased Significantly.
