The root-knot nematode disease was a most serious disease that spread all over the world.It was adaptable and harmful,and it could do harm to about 2000 sorts of plants and spread wildly. The root-knot nematode disease was a serious one in tomato plant, and with the development of dependency and especially the greenhouse ,The harm of root-knot nematode disease became more and more seriously.The damage could reach to 10%-20%,and even to 30%-40%,more serious would have no production.Now there was many methods in practice to provent root-knot nematode disease in tomato,but could not reached a good effection,and it also contaminated environment.So in order to resovle the problem from base,the most effective method is to cultivate resistant breeds.Through this method can reache the most effective result.In this study molecule marker technique was amplied to reseach root-knot nematode resistant gene in tomato.Selecting one RAPD marker linked to root-knot nematode resistant gene.Further more transform it to SCAR marker successfully,and the result became base of clone resistante gene in tomato root-knot disease.It could make tomato itself resist disease,resolve problem in the core.The main results of this study is as followings.1 .In this study tomato resistant breed 04957 and suspecial breed T431 were amplied ,and their hybridize offspring F2 were as the separate groops.Using M.incognita as inoculate material,and using rational inoculate method to mark off risistance and suspeciality gene pool.2.The RAPD molecule marker technique system establishment of the root-knot nematode resistant gene.The root-knot nematode resistant gene in tomato was controlled by single dominant gene .In this study random amplified polymorphic DNAs(RAPD) was employed and applying the method of bulked segregate analysis(BSA) to detect molecular markers linked to root-knot nematode resistant gene in tomato ,Selecting 160 random primers and at last one primer named S60 was selected to further more reseachment.This study identified a new DNA marker S60/1500 that was closely linked to the root-knot nematode resistant gene ,and with the genetic distance of 9.0cM.3.The SCAR molecule marker technique system establishment of the root-knot nematode resistant gene.Based on the nucleotide sequence of S60/1500,a pair of 22nt and 25nt primers was designed...
