Four cattle populations, including purebred Qinchuan cattle(Q) and hybrid offspring cattle, Limousincattle×Qinchuan (LQ), Red Angus×Qinchuan (AQ), German Yellow cattle×Qinchuan(DQ), aged 4 months,totally 164 beef cattle, were used as research populations. The DNA molecular marker techniques, such as PCR-RFLP were applied to evaluate the effects of genotypes on four candidate genes: MYOG, MRF4, H-FABP, CAST genes. The performance traits, including the body weight, body length, heart girth, withers height, height at hip cross, hip width, rump length and ischium width, were associated between genetic marker and performance traits using general linear model (GLM). Clone and analyze of the sequence of bovine MYOG gene; Clone and analyze of the bovine MRF4 gene in silicon; The results were as follows:1 Cloning and Analyze of the Promoter Sequence of Bovine MYOG GeneMYOG gene was investigated to play a core role in regulating myogenisis. According to sequences of MYOG of Homo sapiens, Sos Scrofa and Mus muscles published in GenBank, the homogeneous primers were designed to amplify the MYOG of Bos taurus by performing PCR. The PCR product was ligated to pGEM-T-easy vector, and then E.coil DH5αstrain was transformed. Positive clones were identified by restriction endonuclease, PCR and sequencing. The sequence of MYOG gene of Bos taurus was identified, which showed 86%,78% and 73% homology with that of Sos Scrofa, Homo sapiens and Mus muscles, respectively. It is indicated that the sequence of MYOG gene was comparatively conservative among different species. Cloning and analyzing of MYOG gene made substantial basis for further research on the mechanism of regulation and expression of MYOG gene.2 Cloning and Analyzing of the Bovine MRF4 Gene in SiliconThe cDNA sequence of bovine MRF gene was subjected to Blastn searching against the nr and htgs in NCBI. A working draft sequence (GenBank: AC 138166) was obtained. AC 138166 shows high homology to the cDNA sequence. Blast 2 sequence, Genscan, HMM gene and GENEID algorithms were employed to predict the exons and introns in the homologous region of AC138166.
