Nanoemulsions are thermodynamically stable, isotropic, transparent, dispersed system of surfactant, oil, and water which have characteristic dimensions of 1~100 nm.In this study, in order to prepare pharmaceutical nanoemulsions carrier, we selected Tween80, Span80, ethanol, Isopropyl myristate ( IPM ), Jojoba oil, Vitamin E oil ( VE oil ), and distilled water as ingredient to development pharmaceutical nanoemulsions. the effect of surfactant, cosurfactant, oils, additives, and temperature on nanoemulsion region were examined by using the method of water titration to prepare the pseudoternary phase diagram. HLB value of surfactant, the ration of the surfactant to the cosurfactant, the nature of oils, the change of additives in the water, and the temperature are important in the nanoemulsion system. Then we chosed the system contained of Tween80 / Span80 / ethanol / IPM / Jojoba oil / VE oil / distilled water and Tween80 / Span80 / IPM / Jojoba oil / VE oil / distilled water as the optimization prescriptions according to the pseudoternary phase diagram. We prepared the oil-in-water nanoemulsion for pharmaceutical carrier and dissolved eugenol in the nanoemulsion carrier to 10% eugenol nanomicroemulsion by the optimization preparation process.The method of dyeing with two dyestuffs of Sudan red III and methylene blue was used to determine the nanoemulsion carrier type. The particle size and shape of the nanoemulsion carrier and 10% eugenol nanoemulsion were detected by transmission electron microscope. The stability of the nanoemulsion carrier and 10% eugenol nanoemulsion were tested by high speed centrifugation, constant temperature accelerated test ( temperature at ( 40±2 )℃and relative humidity at ( 75±5 )% ), and normal temperature experiment. The spectrophotometric method was used to determine the concentration eugenol in 10% eugenol nanoemulsion with 282.0 nm wave length at room temperature. Skin allergy and irritation response were detected on rabbits with the nanoemulsion carrier and 10% eugenol nanoemulsion. 10% eugenol nanoemulsion was diluted to 0.5%, 1%, 5% eugenol nanoemulsion solution by distilled water, and used to in vitro acarid-killing experiment and in vivo pharmacodynamic experiment for natural infected acarid rabbits.The result:...
