Objective: The aim of this study is to develop a new type of nanoemulsion preparation fortraditional tea tree oil acaricides,and discuss its stability,safety and efficacy.Methods:(1)By drawing pseudo ternary phase diagram,the tea tree oil acaricides prescription was determined.The type was identified by staining.The morphology was investigated by transmission electron microscopy.The particle size was determined by laser particle size analyzer.The presence of contraindications was determined by high performance liquid chromatography.(2)The stability of tea tree oil acaricidal nanometer preparation was investigated by accelerated test,long-term test and light test.(3)Through thin layer chromatography and high performance liquid chromatography,the ingredient and content analysis methods of the nanoemulsion preparation was established.(4)Skin safety of the nanoemulsion preparation was investigated by skin irritation test.Results:(1)The content of each component of the acaricides was: 1% of eugenol,1% of tea tree oil,1% of thymol,RH-40 9%,3% of 1,2-propanediol and 85% of distilled water.The nanoemulsion preparation is oil-in-oil-type transparent yellow liquid,and the droplets were spherical.(2)In the accelerated test,the drug content of the nanoemulsion was not significantly different from that of 0 months after 6 months.In the long-term test,8 months after the relative content of nano-milk has been reduced,and the valid term is 16 months.(3)The content of eugenol,tea tree oil and thymol were not changed by thin layer chromatography.And they were not changed by high performance liquid chromatography(HPLC).Therefore,there are no incompatibility between eugenol,tea tree oil,and thymol taboo.(4)Rabbit skin irritation test showed that this nanoemulsion preparation has no irritation.(5)In the isolated acaricidal experiment,the mite mortality of this nanoemulsion preparation was significantly higher than that of the acaricidal water(P <0.05).Conclusion: A new type of nanoemulsion preparation for tea tree oil acaricides has been developed,which meets the quality requirements of nanoemulsion preparations,and has good stability,safety and efficacy.
