| Rape (Brassica napus L.) is a kind of oil crops with strong adaptability and economic worth .In our country, the rape is the mainly source of edible oil and genic Male Sterility is very important in producing rape cross bred seed. In recent years, two great changes appear: one is hybridized seed becoming more and more important, the other is common Rape were replaced by especial Rape-whose baleful content is low and beneficial content is high.Molecular marking technique based on DNA is an ideal heredity marking method and along with the appearance of PCR these techniques are developing at very fast speed these years. They will play more and more important role in molecular marker-assisted selection in future. At present, many molecular marking technique are used to find molecular markers. In this study, the dominant genic male sterility Brassica napus L were selected as plant material. Aimed at restorer Gene, RAPD(Random Amplification Polymorphism DNA) and AFLP (Amplified fragment length polymorphisms) were used to look for the markers Linked to the Dominant Genic Male Sterility restorer Gene in Brassica napus L, which are important assistant to breeding and can enhance the efficiency of selection, shorten the cycle of breeding. The results are as follow:1.In 192 pairs of AFLP primers, 7 pairs of primers amplifying bands with obvious polymorphism between the Sterility and the fertile were got: M4/H5,M4/H6,M2/H1,M3/H5,E5/M2,E3/M1,E2/M1, The magnitude of difference fragments are as follow respectively: 470bp, 510bp, 110bp, 150bp, 50bp, 200bp, 810bp.:2. Five of 7 difference fragments were found to be associated with restorer Gene.3. Two of them (M4/H5 & E5/M2) were transformed to SCAR markers... |
