Sequencing Of The Complete Nucleotide Of Porcine Parvovirus SR-1 Strain

Porcine parvovirus causes reproductive failure in swine, fetal mummification abortion and stillbirths.The virus is ubiquitous among swine throughout the world that have been investigated.4 primers were devised from the DNA sequences that have been published in GenBank. To utilize these primers, I used PCR reaction to gain the product. The results revealed that all coding gene and 5'extrem sequences had been amplified from PPV RFDNA cultured in porkling primary kidney cell.Afterward, I used DNAMAN to splice all the sequence, based on the sequence,we can conclude that PPV SR-1 strain is composed of 4877 nucleotides and shares 93.18%,95.67%,95.51%,95.33% identity of nucleotides with that of Kresse strain, NADL-2(5075) strain, NADL-2(5034) strain and NADL-2(4973) strain, respectively. The sequence analysis showed that there were two open reading frames(ORFs) in PPV genome.The left ORF had the coding domains for all of NS gene,The right ORF had the coding domains for all of VP gene.All coding domains interlap, VP2 gene was overlapped with VP1 gene,NS2 gene was overlapped with NS1 gene,NS3 gene crossed NS1 gene and VPl gene.NS1 gene was encoded from ATG to TAA. It was composed of 1989 nucleotides, encoded 662 amino acids.Coding sequence domain of NS2 gene began from the same site as NSl gene. It was divided into 2 parts by an intron of 1472bp, 258bp before the intron encoding 86 amino acids. 225bp following the intron encoding 75 amino acids.Totally, CDS of NS2 gene encoded 161 amino acids.
