Mastitis is a major cause of economic loss to the dairy industry and is also affected by a lot of factors. The breeders focus on how to solve the problem of mastitis by genetic ways. PRKDC gene is important to form inherent immune system. The goal of our research is to verify whether PRKDC gene could be ragrded as a molecular marker to improve the genetic resistance to mastitis.The four fragments of 5 exons of 102 individuals were amplified and then identified the genetype by using the method of SSCP. Among the five fragments ,we found primer2 and primer3 loci exhibited the diversity. There are three genotypes AA, BB, AB on primer2 and its frenqunecies are 0.353, 0.039, 0.608 respectively; three genotypes CC, DD, CD on primer3 and its frenquencies are 0.608, 0.059, 0.333 respectively. The PCR production of primer3 were sequenced 5 individuals, founding A/G variation on 111 locus and T/A variation on 142 locus between different alleles .Least squares analysis showed that herd location, season of calving had significant effects on milk fat rate (P<0.05). Only DD genotype in primer3 had significant effects on milk fat rate (P<0.05). The two diversity loci had no influences on milk production traits. The different genotypes on the two diversity loci of PRKDC gene had no influences on mastitis occus. To compare the homologous on different genera about exon (primer 1), we found that the homologous is 90% between cow and Homo sapiens ,is 86% between cow and Mus musculus.
