Mei flower (Prunus mume Sieb. et Zucc.), as a traditional famous tree, plays an important role in gardening and cultural life in China. Nowadays, the studies on Mei flower are mainly about the cultivation and breeding. And the molecular marker research needs further thoroughness.In order to announce the relative relationship between the Mei flower cultivars, and to lay the technique foundation of molecular marker on Mei flower, this research has optimized the technique of DNA establishing fit for Mei, selected applicable primer for AFLP and cpSSR, and built up 229 DNA fingerprint and 48 cpDNA fingerprint diagrams of Mei flower cultivars.By the AFLP technique, this study built up 229 DNA fingerprints of Mei flower cultivars with the primer E-CAC/M-ACC, The results indicate that the AFLP is good at differentiating the Mei cultivars, especially the cultivars similar in appearance.By the cpSSR technique, this study built up 48 cpDNA fingerprints of Mei flower cultivars with the primer NTCP9 from tobacco cpDNA, which found from 13 primers that were used to amplify the gene pools. The results showed the complexness of relationship and origin of Mei cultivars, and supplied a new way to study the relationship of Mei cultivars.Compared the AFLP and cpSSR technique studied on Mei cultivars, it's showed that, the AFLP is strong at distincting Mei cultivars, and the cpSSR is not fit to differentiating cultivars but can provide a cpDNA heredity information, supplying a new way to study on Mei flower. In the meantime, while adopting the member marking technique to carry on the Mei flower cultivars to authenticate to analyze with relative relation have a necessary usage to have another to lead the thing combination, and need to learn a research with appearance etc. is organic to combine to insure credibility of authenticate the conclusion.
