Lily is perennial bulbous flower and also is famous traditional flower in China.It can be used as food,drug and ornamental plant.There are 90 species all over the world and 64 in china and exists numerous variants, many of them have yet not been used. A great number of precious resources are endangering because of natural disasters, pest and so on.It is urgent to be collected and preserved. lily germplasms from Changbai mountains of northeast area of china, Yunnan province and Taibai mountains of Shan' xi are studied. The bulbs are preserved with tissue culture and author analyzed the genetic diversity using cpSSR and AFLP markers. The results were obtained as follows.1. The 36 species of wild liiy are collected from northeast area of china, Yunnan province. Shan' xi province and other places,such as Beijing ,Fujian, respectively 8,16, 7, 5. The existing status of wild lily is terrible and desiderated for protection at present.2. The AFLP reaction system is builted and optimized through the improvement of traditional CTAB for the high quality DNA and setting disposal of the time of enzymatic cut.Five pairs EcoRI/MseI AFLP primer combination are collected from 60pairs in all.The primers searched 189 alleles.The average number of each primer is 38. EcoRI/Mse1 markers give polymorphic ratio 65.08%. Genetic correlation is generated by AFLP marker between50 species.3. The cpSSR is first attempted in this study, four pairs of cpSSR primer are collected from 11pairs. 12 bands are searched, and the polymorphic ratio is 100%.It is proved that cpSSR was feasible to the analysis of DNA level of lily.AFLP and cpSSR is first introduced in the research for the analysis of genetic diversity. This research is mainly about the protection of lily resources and the analysis of genetic diversity, it is important to the foundation of core germplasm.
